Objective:In recent years,with the rapid development of Chinese medicine cause,market demand of the Radix isatidis is increasing, especially in Gansu Province, its planting area is increasing year by year, but the quality is uneven, so it seriously affected the development of clinical applications and planting herbs industry.In this paper,the research object is estate Radix isatidis in Gansu,for research Radix isatidis resource distribution,cultivation,producing and processing status quo,uneven quality,soil factors and other issues to related research,establishment of evaluation criterion and draft of Radix isatidis in different growth region from Gansu of medicinal soil factors research report,to ensure the clinical safety and effectiveness of using Gansu Radix Isatidis and reasonable standardized planting Radix Isatidis has scientific guiding significance.Methods:By Radix Isatidis herbal research,cultivation,harvesting and processing technonogy,chemical composition and clinical application access,in view of the present condition of Radix Isatidis Gansu resources distribution,planting,processin-g technology of field survey. By comparative study on samples from Gansu province 31 different ares of Radix Isatidis root traits, the observation of microstru-cture, and arginine and acid containing(R, S)- goitrin do TLC study, at the same time,combined with the Chinese Pharmacopoeia2010 Edition under the provision of Radix Isatidis,to detect moisture,toter ash,acid insoluble ash and extract,and using HPLC to(R,S)-goitrin content to determine,the establishment of(R,S)-goitrin to the best scheme according to spring extraction and content determination of polysaccharide composition,has not been stipulated in the Ch.P for determination,determined the polysaccharide extraction and detrction of the best scheme.In addition,the origin of soil physicochemical factors do content detection, correlation and cluster analysis on research,analysis of the impact of soil physical and chemical factors on the medicinal ingredients.Results: Based on the characters of Radix Isatidis in different regions of Gansu(shape,size, surface features, characters of section, texture, smell) research, established the identification standard of Radix Isatis root traits in Gansu.From different habitats of Radix Isatidis moisture content between 4.45%-7.86%;the total ash content between2.83%-6.48%;acid insoluble ash between 0.42%-1.98%;the extract content between25.64%-30.91%;(R,S)-goitrin content between 0.05%-0.42%;the content of polysaccharide between 10.60%-16.22%.Different regions of each component content in Pharmacopoeia standards was within, and higher than that of Pharmacopoeia quality.The moisture content of Radix Isatidis provisional Gansu real estata: was no more than 10%,total ash and acid insoluble ash content examination limited still follow 2010 edition of pharmacopoeia standards,alcohol soluble extractive content consistent with the Pharmacopoeia, shall not be less than 25%,(R,S)-goitrin content: according to the dry matter calculation should not be less than 0.050%,Polysaccharide content should not be less than 8.5%.In addition, the study was found that Radix Isatidis(R,S)-goitrin content and longitude have significant positive correlation(P<0.05), and latitude have negatively correlation(P<0.05),and the polysaccharide content was negatively correlated(P<0.05).Different soil fertility was not the same as the origin of Radix Isatidis in Gansu grow mainly in neutral or slightly alkaline of sandy loam and loess,medicinal parts of Longnan in the read brown soil in the growth,soil organic matter content in the range of 0.5%-3.0%,and most of the soil with high nitrogen,high potassium and low phosphorus.There was also a significant correlation between soil factors,the influence of the quality of medicinal materials to a certain extent, but these soil factors with the same kind of herbal origin of soil physicochemical factors compared with the content was appropriate, conform to the stipulation standard of Ministry of agriculture of the people’s Republic of China,but it still need further optimization of Radix Isatidis for the best growing conditions.According to the Gansu different Radix Isatidis growth to soil fertility in size, 31 different regions of the soil makes cluster analysis,as we can be seen from Figure 5.1 is divided into three categories, fully shows that soil fertility plays an important role in medicine in the process of growth,for the Gansu province to better planning of medicinal herbs planting base and lay the good foundation.Conclusion: By the analysis of the data,we can see that in different places of Gansu Radix Isatidis quality difference,whether it is a routine examination items(moisture,total ash,acid insoluble ash,extract),or the index components [(R,S)-goitrin,polysaccharide] are subject to the altitude,latitude,soil factors affecting,the higher altitude the content of polysaccharide is higher;the higher lower the longitude latitude the more conducive to(R,S)- goitrin contentaccumulation,soil PH and ive total N positive effect of acid insoluble ash content,organic matter and available K content the higher the more disadvantageous to(R,S)-goitrin content accumulation;organic matter and total K of soil PH had a significant negative correlation;total K and available P were positively correlated with organic matter and available P, total N content has positive effect.At the same time,according to the different soil fertility soil classification,to provide a favorable basis for further comprehensive medicinal quality evaluation and reasonable cultivation,and to provide the basic data for the formulation of Gansu Radix Isatidis quality standard draft.
