Objective: By sea cucumber polysaccharide PBMC in vitro cultivation with the human body, to detect T cell surface CD2, the expression of CD28 molecule, for the sea cucumber polysaccharide provide reliable scientific basis for further application, provide theoretical guidance for the development of Marine drugs.Methods:(1) Collecting the West Coast Hospital Respiratory oncology in2013 December ~2014 year in June 30 cases of lung cancer patients, 30 healthy volunteers as control. Extraction of subjects the morning fasting elbow vein blood 4ml, PBMC extraction, respectively with different concentrations of sea cucumber polysaccharide and RPMI-1640 containing10% fetal bovine serum culture medium.(2) To extract the lung cancer patients with peripheral blood PBMC, respecti vely set up blank group, without medicine group, dosing, monitoring of diffe rent concentrations of sea cucumber polysaccharide on cell vitality and the O D value.(3) The sea cucumber polysaccharide and healthy subjects PBMC cultured in vitro, effect on the expression of T cell surface CD2 and CD28 molecules.(4) Flow cytometry is used to test in patients with lung cancer and healthy ch eck-up peripheral blood CD8+CD28+T cells, the expression of CD8+CD28-T cells.(5) The peripheral blood PBMC in vitro cultured sea cucumber polysacchari de and patients with lung cancer, in vitro, were detected CD2 and CD28 expr ession.Results:(1) 0.1, 1, 10, 50, 100μg·m L-1 of sea cucumber polysaccharide on human peripheral blood PBMC in vitro growth condition, we found that With the increase of concentration, cell aggregation was more obvious,especially in 10, 50μg·m L-1, after reaching a certain concentration gathered effect no longer increases; no obvious aggregation phenomenon was observed in blank control group;(2) PBMC proliferation phenomenon began to appear after two hours’ cuiture, as time extended, gathered more and more obvious, the most obvious in 24 hours.(3) Sea cucumber polysaccharide and healthy people before and after peripheral blood PBMC in vitro culture, CD2 and expression of CD28 difference was statistically significant(t= 2.42-6.51, p<0.05)。(4) CD8+CD28+T cells in peripheral blood in patients with lung cancer expression differences statistically significant(T = 4.27, p < 0.001);CD8+CD28-T cells expressing statistically significant difference(T = 4.31, p <0.001).(5)Sea cucumber polysaccharide PBMC and lung cancer patients in vitro,CD2 T cell surface molecule expression training before and after the statistical differences(t=4.29, p<0.001),CD28 molecule expression also statistically difference(t=2.23, p<0.05)。Conclusion: Sea cucumber polysaccharide in certain concentration range can increase peripheral blood proliferation of PBMC, 24 hours of the most obvious; Sea cucumber polysaccharide can adjust CD2 on the surface of the T cells in peripheral blood and the expression of CD28 molecule, indicates that sea cucumber polysaccharide can promote T cell immune function regulation, can improve the body’s cellular immune function; CD8+CD28-T cells in peripheral blood in patients with lung cancer increased significantly,and CD8+CD28+T cells decreased obviously, patients with tumor, low T cell activation function, may result in tumor escape. Sea cucumber polysaccharide has certain anti-tumor activity, is expected to become new type of antitumor drugs and immune modulators.
