Objective To investigate the effects of sea cucumber polysaccharide on PBMC in peripheral blood of patients with lung cancer and normal people,as well as to the T cell surface CD45 RA and CD45 RO molecule expression and it’s clinical significance.Methods Choose 40 healthy persons and 30 clinical diagnosis of lung cancer patients, to collect anticoagulation peripheral blood 4 ml respectively, 1:1 dilution with PBS, centrifugal, extracting PMBC.(1)Respectively given 0.1, 1, 10, 50, 100μg·mL-1 of sea cucumber polysaccharide, to observe PBMC proliferation in vitro after 2, 24, 48 hours culture;(2)firstly to test the espression of CD45 RA and CD45 RO molecules on the T cell with flow cytometer, and then respectively to detect CD45 RA and CD45 RO molecular expression on the surface of PBMC in patients with lung cancer with sea cucumber polysaccharide in vitro before and after.Results(1) 0.1, 1, 10, 50, 100μg·mL-1 of sea cucumber polysaccharide on human peripheral blood PBMC in vitro growth condition, we found that With the increase of concentration, cell aggregation was more obvious,especially in 10, 50μg·mL-1, after reaching a certain concentration gathered effect no longer increases; no obvious aggregation phenomenon was observed in blank control group;(2) PBMC proliferation phenomenon began to appear after two hours’ cuiture, as time extended, gathered more and more obvious, the most obvious in 24 hours.(3) After culturing with sea cucumber polysaccharide for 24 h, the expression of CD45 RA and CD45 RO on healthy persons peripheral blood PBMC has statistically significant difference(t = 2.11~2.23, p<0.05); I t also to lung cancer patients,CD45 RA and CD45 RO, after the CD45 RA and CD45 RO, after culturing with sea cucumber polysaccharide, CD45 RA molecular expression on T cell surface has statistical difference(t = 4.29, p<0.001), while CD45 RO expressed statistically difference(t = 2.23, p < 0.05).Conclusion Sea cucumber polysaccharide in certain concentration range can can enhance the proliferation of peripheral blood PBMC, he most obvious in 24 hours.; for patients with lung cancer, Sea cucumber polysaccharide can adjust the expression of CD45 RO on the surface of the T cells in peripheral blood, it indicates that sea cucumber polysaccharide can be used to improve immunity of cancer patients.
