PARTI:ASSOCIATION BETWEEN TNFAIP3 GENE POLYMORPHISMS AND SUSCEPTIBILITY OF ALLERGIC RHINITIS IN A CHINESE HAN POPULATIONObjective:Polymorphism of the Tumor necrosis factor alpha-inducible protein 3 (TNFAIP3)have recently been reported to be associated with the susceptibility to several immune-related diseases. Allergic rhinitis (AR), a common IgE-mediated inflammatory disease of the nasal mucosa.This study was performed to evaluate the potential association of TNFAIP3 gene polymorphisms and the susceptibility of AR in the Chinese population.Methods:Five single nucleotide polymorphisms (SNPs) of TNFAIP3 were genotyped in 540 AR patients and 524 healthy controls using a polymerase chain reaction-restriction fragment length polymorphism assay (PCR-RFLP). Results:Prevalences of the rs9494885 TT genotype and T allele were found significantly increased in patients with AR. And an conspicuous decreasing prevalence of the CT genotype and C allele were shown in patients with AR. In addition, the CCACA haplotypes were decreased in patients, whereas CTCCA haplotypes were increased.Conclusion:This study suggests that one novel SNPs of TNFAIP3 gene, rs9494885 is associated with susceptibility of AR.PART2:ASSOCIATION STUDY BETWEEN TNFAIP3GENE POLYMORPHISMS AND ASTHMA IN CHINESE CHILDREN, WITH OR WITHOUT ALLERGIC RHINITISObjective:Polymorphisms in the tumor necrosis factor alpha-induced protein 3 (TNFAIP3) gene have recently been associated with susceptibility to several immune-related diseases. Asthma and allergic rhinitis (AR) often coexist and are known as "united airways" inflammatory diseases. This study was performed to evaluate potential associations of TNFAIP3 gene polymorphisms and the susceptibility to asthma, with or without AR, in Chinese children.Methods:Seven single nucleotide polymorphisms (SNPs) of TNFAIP3 were genotyped in 400 asthmatic children and 300 healthy controls using a polymerase chain reaction-restriction fragment length polymorphism assay. Among the asthmatic children,200 had concurrent asthma and AR and the other 200 had isolated asthma.Results:Prevalence of the rs9494885 TT genotype and T allele was significantly increased in children with concurrent asthma and AR, frequencies of the CT genotype and C allele were significantly lower than in controls(P<0.001). In addition, CTCCAAC haplotypes were decreased in children with concurrent asthma and AR, whereas CTCCAAT were increased(P<0.05).Conclusion:To the best of our knowledge, this is the first study to demonstrate an strong association between polymorphisms in TNFAIP3 and children with concurrent asthma and AR. The results suggest that an important association between rs9494885 in a SNP of TNFAIP3 and concurrent asthma and AR.
