Objective:The incidence of laryngeal cancer is about 5% in systemic malignant tumor. Laryngeal cancer is one of the most common head and neck cancer, 95% of its histologic type is squamous cell carcinoma.The prognosis of patients with laryngeal cancer is mainly effected by tumor recurrence and metastasis.Lymph node metastasis is a common cause of death and recurrence in patients with laryngeal cancer, also the most important way of the transfer of laryngeal cancer.Tumor metastasis related gene 1 (MTA1) was originally found in metastases tumor of the rat breast cancer, and it has a promoting effect on breast cancer transfer, so named tumor metastasis related genes.In human malignant tumor such as lung cancer, ovarian cancer, stomach cancer, osteosarcoma,the expression of MTA1 is quantity increased, its high expression of mRNA level is associated with tumor invasion and lymph node metastasis.To observe the effect of metastasis-associated gene 1 (MTA1) regulating the growth and metastasis of laryngeal squamous cell in nude mice using RNA interference technology (shRNA).Methods:Constructed and packaged lentivirus interfering vector of MTA1’s short hairpin RNA (shRNA) and transfected it into the Hep-2 cells, then together scramble shRNA respectively injected into the nude mice (n=5) produced the tumor metastasis model of laryngeal carcinoma.Results:The gene screening showed the lenti viral interfering vector of MTA1 was constructed successfully, and those tumor cells were transplanted and growed successfully in all mice. The size of tumor in mice in MTA1 shRNA tansfected group was obviously smaller compared to the scramble shRNA at the same time point. The result showed the inguinal lumph node were not metastasised in the tumor cells of mice in MAT1 shRNA group.On contrast to the results, the tumor cells were found in the inguinal lymph nodes in the scramble shRNA infected mice. RT-PCR and Western blot nalysis further displayed that the mRNA and protein expression of MTA1, β-catenin, MMP-9, cyclin D in MTA1 shRNA infected mice are obviously decreased compared to the scramble shRNA infected mice.Conclusion:The results showed that the inhibiton of MTA1 gene can down-regulated the growth and mletastasis of laryngeal squamous carcinoma in nude mice,MTA1 may be a potential maker in the diagnosis and gene treatment of laryngeal squamous cell carcinoma.
