Objective To detect the inhibitory effects on B16-F10 melanoma in mice by pulsed electrical stimulation at different low-frequencies and evaluate the effect on their heart structure and functions.Methods The BALB/C mice were established by inoculating with melanoma cell B16-F10. The mice were randomly divided into four groups: the treatment groups exposed to the 1ms,20V/cm, duration of 30min/d,frequencies of lOHz (lOHz group),20Hz (20Hz group)and 25Hz (25Hz group) respectively, and not electrical stimulation to the control group (10 each). Use ECG to record the changes.7 days later, tumor volume and survival rate were generated. The changes in tumor and myocardial morphological were using hematoxylin-eosin (HE) staining. The expression of S-100B protein in tumor was tested by immune histochemistry method. The serum levels of troponin T (cTnT) and creatine kinase isoenzyme (CK-MB) were determined by ELISA.Results After treated with pulsed electrical stimulation for 15 days, the evidence of tumor volume of 10Hz and 20Hz group and 25Hz group (463.026±33.036,248.609±34.620,29.879±15.860mm3, respectively), is significantly less than the control group (2026.989±133.358mm3, P<0.05). Compared with group 20Hz group (67.40%), survival extending rate is significantly increased in group 10Hz (15.47%) and 25 Hz (8.8%); With the increase of frequency, S-100B protein positive expression in turn reduce; In the 25Hz group,arrhythmia score of mice and the serum levels of cTnT or CK-MB were raised (P<0.05); No significant in arrhythmia score of mice the serum levels of cTnT and CK-MB were found among group lOHz and group 20Hz (P>0.05).Conclusion Low-frequency pulsed electrical stimulation have a suppressive effects on the B16F10 melanoma type mice, and the pulsed electric stimulation at 20V/cm,20Hz and 1ms was proved to be the most effective in suppressing the growth of tumor without affecting the normal electrical activity in mice or causing myocardial damage.
