Five extraction methods for propolis were compared in this thesis, and the best one was optimized. Furthermore, the acute toxicity and anti-hypertension of the ethanol extraction of propolis were also studied. The results were as follows.1. The optimized test of the extraction process:Five kinds extraction processes were comp-Ared, incuding traditional maceration extraction(Mac), ultrasound extraction(UE), microwave assisted extraction(MAE,17%power), MAE(40% power) and UE+MAE(17% power). The highest total flavonoid content(0.1770 g/2g) and extraction yield were showed in the method of lower power microwave assisted extraction, which was followed by UE(0.1732 g/2g), Mac (0.1675 g/2g),40% MAE (0.1335 g/2g), UE+MAE (0.0968 g/2g). Then the single factor tests and quadratic regression orthogonal rotary combination method were operated to optimize the process of the microwave assisted extraction. The results of the optimum condition for microwave assisted extraction were as follows:use the ethanol as solvent; microwave cycle value 10 s; extraction times value 2; solid-liquid ratio 1:15; the microwave time value 80 s; and the solvent concentration value 88%.2. The acute toxicity test of the ethanol extraction of propolis:The acute toxicity of the alcohol extraction of propolis was studied by the fixed-dose method. The results of the study were as follow:In the period of 0-14 days after intragastric administration, the mortality of mice was 0; the independent activity of mice were normal; the mouth, nose and ears didn’t appear abnormalities secretion; the eyelids didn’t droop; and the mice were all with clear eye pupil, smooth hair and the normal dung. In addition, the organs didn’t have obvious abnormalities characterization by autopsy, and the organ index didn’t have significant differences, compared with the control group. In conclusion, the ethanol extract of propolis didn’t have severe acute toxicity in normal mice. And because the highest dose for the mice is 5000 mg/kg, which is about 200 times as the human dose, so we can be assured to use the propolis.3. Experimental hypertension mice test:The intervention effect of the drugs (propolis,capto-pril) on L-NNA induced pressor response and other physiological indicators in mice were researched by the method of combinating L-NNA with the drugs. As a result, compared with L-NNA group, the blood pressure of mice were significantly reduced in the treatment group of propolis, but the heart rate of mice were significantly raised(P<0.05). And there was a certain dependence on the dose of the propolis. There was no significantly difference on the blood pressure and the heart rate between the groups of propolis and captopril treatment (P>0.05). At the same time, the L-NNA modeling mice’s cardiac index, the level of the serum’s MDA, AngⅡand ET-1 of treatment groups of the propolis could be significantly reduced (P<0.05). In addition, the L-NNA modeling mice’s cardiac index, the level of the serum’s SOD and NO of treatment groups of the propolis could be significantly raised(P<0.05). In this thesis, the anti-hypertension effect of the combination (propolis and captopril) group was studied, the results were:the combination treatment group was significantly different with the treatment group of the propolis or captopril. But the specific mechanisms and the dose would be studied and discussed further.
