[Objective]To observe the influence of Haoqin Qingdan Decoction on gastric inflammation and COX-2, TNF-a in modle mice with chronic gastritis of spleen and stomach damp heat syndrom caused by Hp infection, in order to reveal its action mechanism of the treatment of Helicobacter pylori associated chronic gastritis,to provide new ideas for the treatment of Helicobacter pylori associated chronic gastritis in TCM.[Method]60 SPF level C57BL/6 mice (male and female each half) were randomly divided into six groups(the control group, model group, Livzon Weisanlian group high-dose Haoqin Qingdan Decoction group, medium-dose group and low-dose group),Each group contains 10 mice. Except control group, the other groups of mice were established the Helicobacter pylori associated chronic gastritis of spleen stomach damp heat syndrom model with composite factors (damp heat environment+fat and sweet diet+Hp infection).After modeling is completed, the control group and the model mice were fed with normal saline group, the other groups were given appropriate medication orally,2 times/day,for 14 days in a row, the mice were killed, HE staining was used to observe the mice gastric mucosa histopathology, immunohistochemistry was used to detect the mice gastric mucosa COX-2, TNF-a expression.[Result]1.The experimental mice gastric pathology:Compared with the model group, high, medium and low dose Haoqin Qingdan Decoction group were able to significantly reduce chronic inflammation score, active inflammation score, degeneration and interstitial edema degree of the gastric mucosa in HAG mice,the difference was statistically significant (P <0.05 or P<0.01).Compared with Livzon Weisanlian group, there is no significant difference in the gastric mucosa chronic inflammation score, degeneration and interstitial edema degree of high, medium and low dose Haoqin Qingdan Decoction group mice, the difference was not statistically significant (P> 0.05), The difference was statistically significant in mice gastric activity inflammation scores of medium and low dose Haoqin Qingdan Decoction(P<0.05).2.The experimental mice gastric mucosa COX-2:increased expression of COX-2 in gastric mucosa of the mice after modeling, the difference was statistically significant(P<0.01); compared with the model group, the treatment group can significantly reduce the expression of COX-2 in gastric mucosa of mice(P<0.01), among which Hao Qin Qing effects of bile Decoction high dose group and Livzon weisanlian group was most obvious(P<0.01).3.The experimental mice gastric mucosa TNF-a:the higher expression of TNF-a in gastric mucosa of the mice after modeling, the difference was statistically significant (P<0.01); compared with the model group, the expression of each treatment group were significantly decreased in mice gastric mucosa (P<0.05 or P<0.01), among which Hao Qin Qing effects of bile Decoction high dose group and Livzon weisanlian group was most obvious(P<0.01).[Conclusion]1.Haoqin Qingdan Decoction can relieve chronic inflammation, active inflammation and degeneration and interstitial edema of the gastric mucosa in mice with Helicobacter pylori Assotiated Gastritis, and there is no dose-dependent; The efficacy of high-dose Haoqin Qingdan Decoction is similar to the Franc Weisanlian.2.Haoqin Qingdan decoction could ruduce the expression of COX-2, TNF-α in gastric mucosa of mice with chronic gastritis of spleen stomach damp heat syndrome infected by Hp, and the high dose group had significant effect, which therapeutic effect close to Livzon weisanlian group.3.The mechanism of Haoqin Qingdan Decoction in the treatment of Helicobacter pylori associated chronic gastritis of spleen stomach damp heat syndrome may be related to anti inflammation and protecting gastric mucosa function by reducing the expression of gastric mucosa COX-2, TNF-α.
