The key to determine the material foundation of traditional Chinese medicine is extraction process, extraction process also affects the forming, stable quality and other aspects of Chinese medicine. In widely used Chinese medicine, the effective ingredient heated is easy to damage or transform adversely account for a certain proportion of Chinese medicine, for this kind of Chinese medicine commonly used extraction method are impregnation and percolation, percolation is the room temperature dynamic extraction, impregnation is room temperature or warm static extraction, both of them can not take into temperature and dynamic account at the same time.Vacuum extraction is a kind of method to achieve low temperature dynamic extraction by reducing pressure, suitable low temperature can ensure maximum dissolution of the effective ingredient not be damaged or transformed adversely,can also reduce dissolution of categories of ingredients; dynamic can ensure enough concentration gradient, which is beneficial to optimize extraction of Chinese medicine. Therefore, this paper studied the suitability of vacuum extraction with sophora as the model drug and achieved the subsequent preparations.In this topic, we had investigated thermal stability of matrine and oxymatrine, and found that matrine and oxymatrine have good stability, but under the influence of other ingredients, oxymatrine heated can convert to matrine. Single factor test and orthogonal design method were used to optimize extraction process parameters:The optimal condition for extraction were as follows:the ratio of liquid to solid 8,extracted for 3 times with temperature was 70℃,1h for each time of vacuum extraction, the ratio of liquid to solid 8, extracted for 3 times, 1h for each time of conventional extraction.After investigation, we have determined the concentration method was vacuum concentration at 60 ℃ and dry method was vacuum drying at 60 ℃.The suitability of vacuum extraction process has been studied, chemical composition of sophora extract by vacuum extraction were different from that by conventional extraction; conductivity, osmotic pressure, Zeta potential and particle size of vacuum extraction extract were different from that of conventional extraction extract, bulk density, compression index, angle of repose and contact angle of vacuum extraction extract were different from that of conventional extraction extract, atmospheric extract were more easily absorb moisture than decompression extract, less critical relative humidity. These studies on the other side reflected that impact of temperature on the extraction of Sophora.Dispensing granule forming process screened type and dosage of materials, type of wetting agent, it had determined the material was dextrin, the ratio of extract to material was 1/1.15, wetting agent was 60% ethanol, sophora dispensing granule 1g was equivalent to the original medicinal 2g. Amplification study in laboratory showed that the process is stable.In the research of quality standards, TLC methods of sophora dispensing granule were established, HPLC was used to content determination of matrin and oxymatrine, and the general rules for inspection of granule preparation were finished. Preliminary studies showed that sophora dispensing granule is basically stable.In this paper, the study on vacuum extraction suitability and dispensing granule forming of Sophora has enriched extraction methods of Chinese medicine, promoted the application of vacuum extraction technology, created the theory of vacuum extraction technology.
