The Research Of JNK Expression In Tong Luo Tang Tai Decoction On Spontaneous Type 2 Diabetic GK Rats With Diabetic Peripheral Neuropathy

Objective:Through the experiments has Yi qi Yang yin Qing re, Huo xue Hua tan Tong luo of Tong Luo Tang Tai decoction research in spontaneous type 2 diabetic GK rats on the DPN effect, aimed at the molecular level, cellular level, the preliminary study of the effect of tongluotangtai decoction on expression of JNK, And this side of the JNK caused by the possible mechanism of DPN to make a preliminary research.Methods:50 7-8-week-old male spontaneous type 2 diabetic GK rats by high fat diet fed for 12 weeks after modeling,according to blood glucose were randomly divided into model group,Tong Luo Tang Tai High-dose group(referred to as high dose group)、Tong Luo Tang Tai Dose group (referred to as dose group)、Tong Luo Tang Tai Low-dose group(referred to as Low-dose group), metformin+Methycobal group (referred to as the western medicine group)with 10 rats in each, Wistar rats with age and sex matched normal group 10, a total of 6 groups.During the experiment, the normal group was fed with normal diet and sterile water, the other 5 groups were fed with high fat diet and sterile water. During the treatment, the normal group and model group were given NS 5.0ml/kg.d by gavage,and Tong Luo Tang Tai High-dose group、dose group、Low-dose group were given Tongluo Tangtai suspension 2.88g/kg.d,1.44g/kg.d,0.72g/kg.d by gavage. the western medicine group was treated with metformin 100mg/kg.d and Mecobalamin 0.2mg/kg.d suspension gavage,1 times per day, all groups were sacrificed 28 days after intragastric administration. To observe the general condition of rats, change detection of rat fasting blood glucose,sciatic nerve conduction velocity, Pathology of sciatic nerve, p-JNK,JNKmRNA and Caspase-3 expression level in sciatic nerve.Results:Compared with the normal group, the blood glucose of rats in the model group increased,sciatic nerve conduction velocity slowed,histopathological damage graved,and up regulate the expression level of p-JNK, JNKmRNA and Caspase-3.After the treatment of tongluotangtai decoction can significantly improve the model in rats with diabetic peripheral neuropathy general, reduce the level of blood glucose, improve sciatic nerve conduction velocity to a certain extent, reduce the pathological damages, the down-regulation of p-JNK and the expression level of JNKmRNA and Caspases-3.Conclusion:Tongluotangtai decoction can down regulate the p-JNK and JNKmRNA in a certain extent expression level, prevent the possible mechanism of diabetic peripheral neuropathy is to inhibit abnormal activation of high glucose stress ind-uced phosphorylation of JNK.Performance in a certain extent,weakened the JNK phosphorylation and gene expression levels,improve the JNK mediated insulin metabolic pathway under high glucose stress caused by environment disorder,reduce cell apoptosis,protect peripheral nerve function, play a protective role of peripheral nerve.
