Background:Vascular endothelial cells(VEC) is a kind of endothelial cells that cover the surface of vascular intimal,which become a barrier between the blood and tissues, multiple kinds of biologically active substances can be synthesized and secreted in that cells.VEC also play an important role in maintain the balance of homeostasis, but in some cases,VEC is susceptible to oxidative damage, which could induce injury.Objective:To investigate the protective effect of blueberry anthocyanin(BA) on endothelial cells and the possible mechanism, from the proliferation activity of cell,cell cycle and superoxide dismutase(SOD) and malondialdehyde(MDA) levels.Methods:This research will choose HUVEC, conventional recovery, training,and passaged,further establish the oxidative stress model of HUVEC.(1)Groups of cell:Usually vitro cultured human umbilical vein endothelial cells(HUVEC) were divided into five groups: control group(Control), ox-LDL group(ox-LDL), ox-LDL+low-dose BA group(ox-LDL+low-dose BA),ox-LDL+middle-dose BA group(ox-LDL+middle-dose BA),ox-LDL+high-dose BA group(ox-LDL+high-dose B A);the concentrations of high, medium and low dose group of BA are respectively 40, 20, 10 mg/ml。(2)Determine BA concentration for experiment: In this part,we use MTT assay to check out cell activity, determine the optimal concentration of the effectively protect concentration of blueberry anthocyanin;(3)Cell cycle were detected by Annexin V-FITC-labeled flow cytometry;(4)Detect the level of SOD and MDA.(5)Detect the intracellular ROS level.Results:Oxidative stress of HUVEC was significantly increased after the stimulus of ox-LDL,Compared with ox-LDL group, SOD level of the ox-LDL+BBA Group was significantly increased, the level of MDA decreased, the intracellular ROS level also in a downward trend.Conclusions:1.Ox-LDL can induce oxidative stress of cultured endothelial cells.2.Ox-LDL induced HUVEC oxidative stress, may be related to the imbalance level of SOD and MDA.3.BA has a kind of inhibition effect of HUVEC oxidative stress,which can protect vascular endothelial cells and plays a role in anti-atherosclerosis.
