Nelumbinis Plumula is the dried spire and radicle of the mature seed of Nelumbo nucifera Gaertn of Nymphaeaceae, which has been used to treat annoyance, insomnia, spermatorrhea, thirst and hypertension. The primary active ingredients in Nelumbinis Plumula are alkaloids and flavonoids. Pharmacological studies have shown that it has the function of anti-tumor, cardiovascular protection, oxidation resistance, inhibiting liver fibrosis, hpyerglycemic, antibacterial and anti-inflammatory, etc.In this paper, Nelumbinis Plumula was mercerated with 80% alcohol at room temperature, and the ethanol solution was vacuum evaporated to recover the solvent until no-alcohol. Then the concentrated solution of total extraction of Nelumbinis Plumula was obtained. And then the chemical components were analysed by using solvent extraction, silica gel column chromatography, high preparative performance liquid chromatography and recrystallizing, etc. Fourteen monomeric compounds were obtained, and the structures were identified by the physical and chemical properties and spectra analysis(13C NMR, 1H NMR, etc). They were: N-methycoclaurine(1), neferine(2), isoliensinine(3), liensinine(4), rutin(5), galuteoline(6), hyperin(7), vitexin(8), kaempferol(9), chrysoeiml-7-O-[2’’-O-β-D-glucopyranosyl]-β-D-Glucopyranoside(10), 1-chlorohexadecane(11), β-sitosterin(12), daucosterol(13), mannitol(14). Among them, Compound 1, 9, 10 and 14 were discovered from Nelumbinis Plumula for the first time.The anti-tumor activitives of total alkaloid and alcohol extractive of Nelumbinis Plumula were observed by MTT method. The results indicated that total alkaloid could restrain proliferation of human neuroblastoma SH-SY5 Y and human glioma U251. Furthermore, the inhibiting effects were positively ralated with the concentration of total alkaloid. This study provided the theoretical basis for the further development and use of Nelumbinis Plumula.
