Plumula Nelumbinis,the green embryo of seed of plant Nelumbo nucifera Gaertn,was first recorded in "Shixing bencao",and now was embodied in Chinese Pharmacopoeia(Edition 2000).As a well-known herb,Plumula Nelumbinis was widely used in traditional Chinese Medicine(TCM) and have bitter taste with cold proporty.It was usually used in high fever,insomnia,spermatorrhea,haematemesis.In modern research, alkaloids such as liensinine,isoliensinine,neferine and lotusine can better debase blood pressure,antagonize arrhythmia,antagonize insulin resistance,etc.It has wide therapy foreground in desease of blood vessel in heart and brain.Firstly,I summarized the chemical components of Plumula Nelumbinis with on the aids of references.Besides,I summarized the chemical components and pharmacological properties of non-phenolic alkaloids from Plumula Nelumbinis,which provided the foundation of the study.Secondly,I studied on the non-phenolic alkaloids and non-alkaloidal constituents of Plumula Nelumbinis.Compounds were separated by solvent extraction,polyamide and silica gel column chromatography.Their structures were elucidated by spectroscopic analysis.Eight compounds were isolated from Plumula Nelumbinis.They were identified as: palmitic acid(Ⅰ),ethyl palmitate(Ⅱ),glycerol tripalmtate(Ⅲ),palmitamide(Ⅳ),β-sitosterol(Ⅴ),daucosterol(Ⅵ),N-methylcorydaldine(Ⅶ),nuciferine(Ⅷ),lysicamine (Ⅸ),N-methoxycepharadione B(Ⅹ).CompoundⅡ,Ⅲ,Ⅳ,Ⅶ,ⅨandⅩwere obtained from this herb for the first time.Thirdly,I observed the antihypertensive and reverse effects of two kinds of phenolic alkaloids extacted from Plumula Nelumbinis.liensinine perchlorate(Lien) and lotusine(Lot) on left ventricular hypertrophy of renovascular hypertensive rats(RHR).RHR were formed by undergoing the two-kidney,one-clip(2K1C) procedure,and formed left ventricular hypertrophy(LVH) after 4 weeks of clipping.They were given Lien 10 mg/kg·day,20 mg/kg·day and Lot 10 mg/kg·day,20 mg/kg·day p.o.for 10 weeks,and the systolic pressure were determined before and after treatment.The improvement of LVH of the two alkaloids were assessed by observing the ratio of left ventricular weight to body weight (LVW/BW) in RHR.The parameters of the left ventricular structure were also determined. The systolic pressure were decreased obviously(P<0.01 or P<0.05) after 4 weeks of treatment;the LVW/BW and the thickness of septa interventriculare(SI) and left ventricular wall(LVW) of RHR were all decreased significantly(P<0.01 or P<0.05).We can concluded that Lien and Lot can decrease blood pressure and have a good effect on left ventricular hypertrophy of RHR.Finally,the effects of four kinds of phenolic alkaloids from Nelumbo nucifera (liensinine,isoliensinine,neferine,lotusine) on the functions of HUVECs were performed in this study.The effects of four kinds of alkaloids on HUVEC injury induced by H2O2 are observed by determination of MTT,NO and NOS.The results and conclusion:Four kinds of alkaloids have the protective effect on HUVEC injury induced by H2O2.The mechanisms of four alkaloids to the protective effect on HUVEC injury by H2O2 were not the same.
