Experimental Study On Alzheimer Disease Model-rats’ Ethology And Oxidation Resistance Effects Of Decoction Made Up With Renshen, Sanqi And Bingpian

Purpose: Observe decoction made up with Renshen, Sanqi, Bingpian’s(RSB decoction) influence to AD rats’ ethology and oxidation resistance.Methods: Select 60 normal rats with water maze and divide them randomly into Group A( blank group), Group B(disguised operation group), Group C( model group), Group D( Piracetam group) and Group E( RSB Decoction), with 10 rats in each group. We keep Group A intact and inject salineinto hippocampus and enterocoelia in Group B’s rats. With Groups C, D, and E, we give D-Gal and Aβ1-42 together into rats by intraperitoneal injection for 42 days to copy AD model. Groups D and E respectively gastric with different drugs, rats in Group D are treated with piracetam, which has proven to be an effective drug for AD, and rats in Group E are treated with RSB Decoction with the dosage of 0.25g·(kg/d)-1 for 21 consecutive days. After the drug treatment, all rats are tested with the water maze. Before anatomy the rats, all rats are fasted for one night. Collect blood samples from abdominal aorta and measure the content of SOD, MDA, IL-6, T-CHO with ELISA. Fixing the rats’ brain through the heart perfusion with 4% par formaldehyde in 4℃, then speedily conduct a craniotomy operation to pick off hippocampuses on ice cubes and make morphological observation. Picking off their hippocampuses on ice cubes, without fixing, we add normal saline, homogenate it, obtain the supernate, and measure the scontent of Aβ in rats’ hippocampuses with ELISA.Results: 1. In ethology terms, compared with the normal and disguise groups, model group’s escape latency is significantly increased; Compared with the model group, the drugged groups’(D, E) escape latency are significantly decreased. Compared with the normal disguise groups, model group has longest dwell time in target quadrant and lowest times in crossing the platform and there is no difference between group D and E. The normal group and disguise group chose orthoscopic and tendency as the search strategy. Model group rats use edge type and random type, and some individual chose tendency type. Rats in group D and E use more methods of tendency compared with edge and random methods. D group and E group,no statistically significant difference. 2. In biochemical terms for blood, compared with the normal and disguise groups, modelgroup’s MDA content in serum is significantly increased, SOD content is decreased. Compared with model group, drugged groups’(D, E) MDA content in serum are significantly decreased and and there is no difference between group D and E. Compared with the normal and disguise groups, model group’s IL-6 content in serum is significantly increased, and compared with model group, drugged groups’(D, E) IL-6’s content in serum are significantly decreased and and there is difference between group D and E. Compared with the normal and disguise groups, model group’s Aβ and T-CHO content in serum are significantly increased, and compared with model group, drugged groups’(D, E) Aβ and T-CHO content in serum are significantly decreased. There is no difference between group D and E for Aβ, but group D is better than group E for T-CHO. 3. From the tissue pathogenesis terms, the normal and disguise groups’ hippocampuses area CA1 have complete structure. The neurons are in alignment and compact, layers are coherent, nucleus of the neurons are round, big and regular, and the nucleolus of the nucleus are clear. Model group’s hippocampuses area CA1 are in a mess and lost. The cells are swollen, neurons are dark dyed and in pyknosis. The neurons present a state of granular-vacuolar degeneration widely. Piracetam group and RSB groups’ neurons in hippocampuses area CA1 have improved than model group, the neurons are loose and in a mess and layers decreased than blank group. The swollen, dark dyed, pyknosis and granular-vacuolar degeneration condition decreased than model group. Piracetam group and RSB group is almost the same in the pathogenesis change.Conclusion: 1.With the methods of combination of D-Gal and Aβ1-42 together injection into rats, the abilities of learning, memory and antioxidant decreased, this shows the combination methods in copying AD can greatly stimulate the character of AD patients’ ethology and oxidation resistance. 2. RSB decoction can improve the content of SOD, lower the content of MDA and IL-6, this suggests it has the function in improving the ability of antioxidant and it is better than Piracetam. 3. RSB decoction can lower the content of Aβ in hippocampuses, decrease the pathological products, and it is similar with Piracetam. RSB decoction can lower the content of T-CHO, decrease the dangerous factors of launching AD, but it’s worse than Piracetam.4. RSB decoction can obviously improve the pathogenesis condition of the hippocampus in AD model rats.
