Objective:Methylation-specific polymerase chain reaction (MSP) was used to investigate the methylation status of the CpG islands of in the promoter regions of 14-3-3σ,14-3-3ζ and 14-3-3ε subtypes in the ovarian endometriosis cyst, with the aims of detecting the roles of methylation status of these subtypes in the pathogenesis of ovarian endometriosis cyst.Methods:To choose the endomeriosis specimens of the ovarian endometriosis cyst that has significant differences or not parallel expression between mRNA and proteins, and the endomeriosis specimens of the benign ovarian cyst (simple cyst or teratoma) by the pathological diagnosis as the normal group. Separated, purified and cultured the stromal cells, then detected the methylation status of 14-3-3 a,14-3-3ζ and 14-3-3ε subtypes in the normal group and the control group by MSP.Results:Compared with the stromal cells of the normal group, the endometriosis mesenchymal cells of the ovarian endometriosis cyst:14-3-3σ subtypes exists the methylation in the ovarian endometriosis cyst; 14-3-3ζ subtypes exists to methylation; 14-3-3 s subtypes have no obvious difference in the methylation.Conclusions:The abnormal methylation of 14-3-3σ and ζ subtypes in the ectopic endometrium tissues ovarian endometriosis cyst which suggested the two subtypes of 14-3-3 proteins may be involved in the development of the ovarian endometriosis cyst.
