Physalis alkekengi L., plant of Solanaceae, mainly used for the treatment of sore throat, hoarseness, cough, oliguresis and so on.The edible berries with rich nutrition are wrapped up in the calyces that like a lantern.People in the North always hang the fruits under the eaves,and eat them after frost.What's more, modern pharmacologic research has showed that the fruits of Physalis alkekengi L. possess the activity of anti-hyperglycemic.Firstly,HPLC-DAD-ESI-MS2 was used to analyze the active fractions?ethyl acetate extract and n-butyl alcohol extract? from the fruits of Physalis alkekengi L..And the possible structure of 9 chemical constituents in the active fractions were identified.Secondly,7 compouds were isolated from the ethyl acetate extracts and n-butyl alcohol extracts of Physalis alkekengi L..by using the silica gel column chromatography and gel column chromatography etc..6 compouds were identified by physicochemical properties and spectroscopic data,they are ferulic acid,luteolin-7-O-glucoside,quercetin-3-O-glucoside, kaempferol, trans-p-coumaric acid and quercetin.Thirdly,this paper investigate the anti-hyperglycemic and antioxidant activity of the extraction and the monomer components that separated from the fruits of Physalis alkekengi L..The inhibitory effect on two resources ?-glycosidase were used to evaluate the anti-hyperglycemic activity. The result indicates that the inhibitory effect of n-butyl alcohol extract on ?-glycosidase from small intestines of rats and drugstore was better than other extraction and acarbose. And the IC50 values were 0.321, 0.161mg/ml, respectively. At the same time, the scavenging effect on ABTS+· of each extraction and compounds were evaluated.In which the IC50 values of ferulic acid?0.011mg/ml?is approach to Vc?0.012mg/ml?.Then,this paper studies the effect of ethanol concentration, material-solution ratio,extraction time and extraction times with the total flavonoids and total phenols as the index. The optimum extraction process for the fruits of Physalis alkekengi L.is confirmed by orthogonal test as follows: 85% ethanol, liquid material ratio1:20,extracting time1.0h, extracting times 2.Lastly,the HPLC was used to determine the content of 3 compounds?ferulic acid,luteolin-7-O- glucoside and quercetin? in the fruits of Physalis alkekengi L.,which were obtained from 13 different areas.The results showed that the highest value and the lowest content of ferulic acid were obtained from Jiaohe(0.121 mg·g-1)and Shulan(0.012mg·g-1),respectively; the highest value of luteolin-7-O-glucoside were obtained from Zhuchengzi(0.098 mg·g-1),and the minimum was obtained Shulan(0.0065mg·g-1); the highest value and the lowest content of quercetin were got from Helong(0.0608 mg·g-1)and Tonghua(0.0128 mg·g-1),respectively. The figerprints of the fruits of Physalis alkekengi L.were established,16 common peak were choosed among the fingerprints.What's more,the similarity of 13 different areas of Jilin province were greater than 0.9.
