| Objective: The aim of this study was to investigate the level of motilin in serum and the expression of i NOS in the colon of slow transit constipation rats, and to explore their mechanisms.Methods: 60 SD rats were randomly divided into two groups:control group and model group. Rats in the model group were intragastric administration of compound diphenoxylate, while the blank group with distilled water. After the success of the model, rats in model group and blank group were given distilled water to fill the stomach, and the other groups were given corresponding drug treatment. After the end of treatment, specimens were collected. The morphological changes in the colon were observed, the level of MTL in serum was detected, and the expression of i NOS in the colon of the rats was observed.Results: Compared with the blank group, the number of colonic ganglion cells in the model group was significantly reduced, vacuole degeneration of ganglion cells in the colon was detected, the fiber was slightly swelling, congestion and edema in submucosa were saw occasionally; compared with the model group, the morphology of colonic ganglion cells was basicly normal and the number of nerve cells were not significantly reducd in Cisapride group and high dose group, while vacuole degeneration was found in the two group. Compared with model group, the content of MTL in serum significantly increased in Cisapride group and traditional Chinese medicine of low, medium and high dose group, the difference is statistically significant(P<0.05).Compared with the model group, the expression of i NOS in the colon decreased in Cisapride group, the difference is statistically significant(P<0.01),the expression of i NOS in colon decreased in traditional Chinese medicine high dose group, the difference is statistically significant(P < 0.05).Conclusion: Xuan Fei Bu Shen Yang Yin decoction may improve the symptoms of slow transit constipation by improving the lesion of intestinal ganglion, increasing the content of motilin in serum and reducing the expression of i NOS in the colon. |
