The Study On Characteristics Of Absorption Panax Notoginsenosides On Resins

Object:Panax Notoginseng is the roots and tubers of panax notogising(Bur K.)F.H Chen(Araliaceae),which pocess hemostasis, blood stasis and swelling analgesic effect. Its main component is radix notoginsen saponin, Panax Notoginsenosides(PNGs) play important physiopathological and therapeutic roles in heart disease, angina pectoris and cerebrovascular disease. It has been used widely in clinic.This paper evaluates original extraction process of PNGs, several aspects contentare included in this paper as folowing:The influence of concentration, temperature and loading volume and elution volume and using time of decolorizing resin on adsorbtion ability, The ultimate aim is to consummate craft standard of separation and purification and compatible with content requirements in elemene injection of Chinese Pharmacopoeia 2015 version.Methods:1. Establishing a method to determinate the content by HPLC and validate the methodology.2. Effects of concentration of extract solution :using eaual amouts of resin and place it in diffrent concentration of PNGs. According to the adsorption rate of resin, evaluating The influence of concentration on adsorption ability of resin.3. Affection of temperature on adsorption capacity:Puting equal amounts of resin in different temperatures.At 0, 25, 40, 50 ℃, then detecting combining rates respectively.4. Dynamic adsorption experiments of resin:on the one hand, Making dynamic adsorption curve on the basis of receiving liquid were taken at diffrent volume.From the diagram one can determine loading capacity of the resin;on the other hand, judging elute volume acorrding to the site of changes of dynamic adsorption curve and Industrial needs.5. Study on the effects of decolorization in the using times of decolorizing resin: Plot of the bar chart with the using times of decolorizing resin as the abscissa and the Color contrast as the ordinate. Acorrding to third wave of small-scale tests by the experimental results of the previous chapters,regarding the pharmacopoeia 2010 edition as standard.Results:1. The paper compares the isothermal adsorption curves of PNGs to curves of I-V gas adsorption,The results show that the curves of Rd is similar with curves of adsorption II,the rest of Notoginsenoside is similar with V,Based on the above results, it is concluded the adsorption of resin is monomolecular adsorption initially.After saturation,the adsorption of second and third is stated on the resin surface.After reaching a certain concentration,PNGs is precipitated in the solution On the surface of resin.from the curve of the adsorption,we found that PNGs is on continuous adsorption.2. Affection of temperature on adsorption capacity:At 0, 25, 40,50 ℃,the adsorption capacity of the resin was better at 50 ℃, so the adsorption process of H resin was endothermic.3. Dynamic adsorption experiments of resin: The extraction of 0.2 g material/ml was adsorbed in 1.3 times of thecolumn volume at a speed of one column volume per hour. extraction was eluted by 70% ethanol of 3 times with the column volume.4. Study on the effects of decolorization in the using times of decolorizing resin: The color decreases as the using times of decolorizing resin increases,the fourteenth times is completely invalidated.5. The elution rate was above 86%, The average content of the radix notoginsen saponin was 85.12%, the content of total saponins were85.2%,85.4% and 84.9%respectively, which met the requirements of Pharmacoeia except the third.6. The Quality analysis of three PNGs of water injection was reviewied,The adverse reaction of traditional Chinese medicine injection status, clinical application status, existing problems and ways of improvement.Conclusion:The adsorption characteristics of H resin at different concentration and different temperature were studied by Characteristics of Absorption Panax Notoginsenosides.On the basis of the original process,The paper provides the technological parameters for the sample volume and alcohol elution of radix notoginseng saponin and standardize the adsorption process and Desorption process.The whole process is simple and easy to operate,both of dosage of eluting agent and alternatives cost were reduced,The content of PNGs was in accordance with Chinese pharmacopoeia 2010 edition.finally The article give some opinions on separation and purification of resin and the have a certain impac for pilot test procedure.
