Radix Astragali has a very wide range of pharmacological effects,and its Immunomodulatory effect has been commonly used in clinical practice.As a popular research direction in recent years,spectrum-effect relationship of Traditional Chinese medicine(TCM)could combine its chemical components with the efficacy,which could evaluate the quality of TCM scientifically.At present,the measured efficacy indicator is usually single in the research of immunocompetence spectrum-effect relationship of TCM.However,the immunological activity of TCM is manifested in many aspects.Therefore,we explored the Immunomodulatory effect of different Radix Astragali extracts on normal and immunosuppressed mice,and discussed the spectrum-effect relationship of the best part in Radix Astragali.The gray correlation analysis was used to combine fingerprints with multiple immunological indicators,and the active components were confirmed.This paper is mainly divided into the following two parts:The first part mainly studied the pharmacological activities of different Radix Astragali extracts on immunomodulatory effect.After oral administration of different Radix Astragali extracts on normal and immunosuppressed mice,phagocytic index,delayed type hypersensitivity(DTH)degree,leukocyte count,organs index and the level of cytokines IFN-?,IL-4(ELISAs)were investigated.By the comprehensive comparison of different extracts,it is found that supernatant extracts of water decoction are the best part of immunological activity.The immunomodulatory effect of different source of Radix Astragali supernatant extracts on immunosuppressed mice was studied.It is found that 10 batch of Radix Astragali supernatant extracts can improve immunological function on immunosuppressed mice to varying degrees(P<0.01,P<0.05).Shanxi Hunyuan,Gansu Lianfeng,Gansu Shuangquan and Shanxi Weinan these four sources of Radix Astragali exerts a better effect in Immunoenhancement.The second part mainly studied the immunocompetence spectrum-effect relationship of Radix Astragali supernatant extracts.The HPLC-DAD and HPLC-ELSD fingerprints of supernatant extracts were established.19 common peaks were determined and 11 compounds were identified.The gray correlation analysis was used to correlate fingerprints with immunological indicators.The results showed that the chemical component of Calycosin-7-O-?-D-glucoside,2'-Hydroxy-3,4'-dimethoxyisoflavan-7-O-?-D-glucoside,Calycosin-3-O-?-D-glucoside,Ononin,Astragaloside IV,Astragaloside III,Astragaloside II were found to have a great contribution to the immunological function.In this paper,we selected the best part of immunoenhancement effect in Radix Astragali,and established the fingerprints of Radix Astragali supernatant extracts based on the immunoenhancement effect,and revealed the therapeutic basis of immunocompetence in Radix Astragali.
