By carding the related documents of liver-spleep-harmonizing and miscellaneous di-seases,thinking the pathological changes of qiand blood are the pathological basis of miscellaneous disease,as well as liver and spleen two dirty are the core of generate an-g run the qi ang blood, moreover viscera and channels as the core of syndrome diffe-rentiation,treatment of miscellaneous diseases attaches great importance to the different-iation,and differentiation of liver and spleen two dirty as the core. Therefore liver spl-eep-harmonizing for the treatment of miscellaneous diseases have a high clinical signi-ficance. In recent years, the treating thinking of miscellaneous diseases also used in c-linical and experimental study of mammary glands in.This topic on the basis of probi-ng liver and spleen in the role of mammary glands and explore dangguishaoyaosan tr-eatment mechanism,conducting experimental study of supplemented dangguishaoyaosan to apoptosis of breast epithella cells in rats on mammary gland hyperplasia and P53 gene expression.Objective:Through the animal experiment to observe the effect of supplemented dangguishao-yaos to apoptosis of breast epithella cells in rats on mammary gland hyperplasia and P53 gene expression to evaluate the main efficancy of supplemented dangguishaoyaos, provide a scientific basis for chinese medicine from liver-spleep-harmonizing on the tr-eatment of breast hyperplasia and breast cancer prevention.Methods:Choose 40 SD female henlthy not pregnant rats,free access to water by eating,nat-ural light,adapt to the environment after a week,were randomly divided into blank co-ntrol group contained 8 rats and test group contained 32 rats.Excepting the bland con-trol group,there test group used the method that al.sex hormone joint msked model fo-r making mammary gland hyperplasia pathology model.Then the experimental group w-ere randomly divided into model group 1,model group 2,supplemented dangguishaoya-osan group,rupixiao groups, each contained 8 rats.While execute model group l,by ob-serving the model 1 set of breast tissue under a microscope to determine pathological model building of mammary gland hyperplasia in rats.After making model,the treatme- nt group were served irrigation supplemented dangguishaoyao san,while the treatment c-ontrol group were fed with rupixiao solution,the model 2 group and blank control gr-oup were poured nomal saline.After filling the stomach 30 days.All experimental rats were sacrificed and specimened rat mammary gland tissues,to observe the change of n-ipplediameter and height,observe breast tissue pathological,using tunnel test rats of eac-h group breast epithelial ceel apoptosia and using per method to detect the expression of P53 gene.Results:①Comparison of the diameter and height of the nippleModel group(1,2),rupixiao group,nipple diameter supplemented dangguishaoyaosan group rats were greater than the height of the blank group,P<0.01, model group 1 and group 2 rat nipple diameter,height was not statistically significant,P>0.05.Rupixiao gro-up rats and supplemented dangguishaoyaosan nipple diameter and height were not stat-istically significant,P>0.05;statistically significant;rupixiao group and large group supple-mented dangguishaoyaosan nipple diameter,height less than the model groou(1,2),P<0.0 1.statistically significant.②Comparison of the breast epithelial cell AIThe breast epithelial cell AI of each experimental group were lower than the Bla-nk group,P<0.01, statistically significant.the breast epithelial cell AI of model group (1, 2) was the lowest, two group were not statistically significant,P>0.05;the breast epithe-lial cell AI of supplemented dangguishaoyaosan and rupixiao groups were higher than the model group (1,2),P<0.01,statistically significant,but between the two groups were no significant difference,(P>0.05).③Comparison of P53 gene expressionThe P53 gene expression of each experimental group were higher than blank grou-p,P<0.01,statistically significant;the P53 gene expression of model group (1,2) were no-t statistically significant,P>0.05;the P53 gene expression of supplemented dangguishao-yaosan and rupixiao groups were lower than the model group (1,2),P<0.01,statistically significant;the P53 gene expression of supplemented dangguishaoyaosan was lower tha-nrupixiao group,P<0.01,statistically significant.Conxlusion:Dangguishaoyaosan supplemented can effectively reduce the diameter and height o-f the nipple,at the same time can be significantly improved the inhibition degree of p-rohyperplasia of mammary gland,its mechanism may be through regulate liver,spleen a-nd huayu sanjie ang adjust the balance of whole blood to reduce the expression of t-he P53 gene,thereby improve the status of local histology to cure proliferation of ma-mmary gland ang prevent cancerous.
