Effect Of Taohechengqi Decoction On Micro Inflammatory State Of Chronic Renal Failure Rats

Purpose:Explore effect of Taohechengqi Decoction on micro inflammatory state of CRF rats, so as to clarify the Taohechengqi Decoction in the treatment of CRF micro inflammatory state mechanism.Method:1.To establish the model of chronic renal failure rat:60 male SD rats,7 weeks old, weighing 180-200g, adaptive feeding for 1 week after repurchase, were randomly assigned to normal group, sham operation group, model group, control group, treatment group,with 12 rats in each. In model group, control group and treatment group were treated with the 5/6 nephrectomy in animal models; sham operation group only double kidney is membrane peeling; normal group received no surgery.2.After successful modeling,4 weeks after operation, the animals were administered once a day in the morning:treatment group given Taohechengqi Decoction gavage; while the control group was treated by Niaoduqing gavage; normal group, sham operation group and model group were given normal saline gavage.3.To drug after the 57 day, urine and the abdominal aorta blood was collected, detection of renal function (Scr, BUN), blood (Hb, RBC),24h urinary protein (sulfosalicylic acid), serum and urine β2-MG (radioimmunoassay), urine NAG (chemical colorimetric method), urinary RBP (ELISA method), serum cytokine levels of CRP, TNF-a, IL-6, ALB (ELISA method), observing the pathological changes of renal tissue by electron microscopy and light microscopy.Result:1.Model group:24h urinary protein significantly increased, Scr, Bun significantly increased, Hb, RBC significantly decreased, β2-MG, NAG, RBP significantly increased, CRP, TNF-a, IL-6 significantly increased, ALB significantly decreased, renal interstitial fibrosis. (P<0.05)2.Control group and treatment group:compared with the model group, control group and treatment group reduce 24h urine protein, Scr, BUN, β2-MG, NAG and RBP, increase Hb, RBC, reduce the micro inflammatory factor CRP, TNF-a, IL-6, increase ALB and the degree of renal interstitial fibrosis reduced. (P< 0.05)Conclusion:1.Taohechengqi Decoction can protect and improve renal function and blood routine index, glomerular and tubular function, delaying the process of renal fibrosis;2.Taohechengqi Decoction can reduce the content of CRP, TNF-a, IL-6, increase the content of ALB, delaying progression of chronic renal failure.
