Objective The expression of TGF-?1 and MCP-1 in the glomeruli and renal tubules ofrenal tissue induced by adenine-induced CRF model rats were observed.The levels of BUN,Scr,P,Ca and the expression of IL-6,TNF-? in serum of rats were studied.The effects of Kaijie Decoction on chronic renal failure rats were studied.Method On the basis of pre-experiment,adenosine intragastric administration of CRF model rats was performed.SPF SD rats were randomly divided into 6groups accdingto body weight:the control group and the model group were treated with normal alin(10ml·kg-1·d-1),low-dose group(3.1g·kg-1·d-1),high dose group(12.6g·kg-1·d-1),possive control group(2.63g·kg-1·d-1),gavage 1 times.After 30 days of treatment,the rats were anesthetized with chloral hydrate intraperitoneal injection(3ml/kg).The rats were sacrificed and the apex was exposed.The heart blood(5ml)was extracted with a onetime vacuum blood collection tube.The kidneys were taken and the kidney size,texture and color were observed,clean,quickly placed in 4% paraformaldehyde liquid fixed.The pathological changes of glomeruli and renal tubules were observed by HE staining.The contents of BUN,Scr,P and Ca were detected by immunohistochemistry.The expressionof TGF-?1 and MCP-1 in renal tissue was detected by ELISA.Serum IL-6,T NF-? content.Results 1.Kaijie Decoction can significantly improve the general situation of CRF rats,in each group of rats during drug intervention,weekly weight changes recorded.Compared with the other five groups,the weight of the rats in the control group was s-ignificantly higher than that in the other five groups.Compared with the other group s,the weight of the rats in the model group was significantly lower than that in theother groups.The level of body weight gain in rats was significantly the same as that inure-mia group.The body weight of rats in high dose group was not obvious.HE staining showed that a large number of dark brown crystals were deposited in the renal tubules,and the renal tubular structure was destroyed.A large number of inflammat oryfactors were infiltrated,the glomeruli were atrophy and the proportion of the balloon was increased.Dose group and uremic clear group compared with the model group,reduced renal tubular damage,showing renal tubular disorder,renal interstitial inflammatory cell infiltration,glomerular no obvious atrophy and sclerosis performance(P>0.05).2.The levels of BUN,Scr and P in the serum of the rats in the model group were significantly higher than those in the blank control group(P<0.01).After 30 days After treatment,the contents of BUN,Scr and P in the low,middle and high dose group and the urine group were significantly lower than those in the model group(P<0.05).Compared with uremicqing treatment group,the effect of low and middle dose group was significantly higher than that of uremia group(P<0.05).There was no significant difference between high dose group and Niaoduqing group.There was no significant difference in the content of Ca in each group(P>0.05).3.Kaijie Decoction can effectively reduce the levels of IL-6 and TNF-? in serum of CRF model rats.ELISA test result shows: compared with blank group,the content of IL-6 and TNF-? increased obviously in serum of CRF model rats,the difference is significant(P<0.01).Compared with model group,the levels of IL-6 and TNF-? in theserum of the low,medium and high dose treatment group and Niaoduqing group were significantly lower than those in the model group(P<0.05).The levels of IL-6 and TNF-? in the middle dose group were significantly lower than those in the control group(P<0.05).The low dose group and the high dose treatment group had no significant difference Statistically significant(P>0.05).4.The expression of TGF-?1 and MCP-1 in renal tissue of each group was mainlyobserved in the renal tubular epithelial cells,and the cytoplasm was mainly stained expression;MCP-1 mainly in renal tubular expression,glomerular also a small amount of expression.Compared with the blank group,the expression of TGF-?1 and MCP-1in the model group was significantly higher than that in the blank group(P<0.05),and the expression of TGF-?1 and MCP-1 in the blank control group was significant-ly higher than that in the blank group Compared with the model group,the levels of TGF-?1 and MCP-1 in the low,middle and high dose group and uremia group were significantly lower than those in the model group,and the expression of TGF-?1 and MCP-1 was significantly higher in renal tubular epithelial cells Compared with the urine group,the expression of TGF-?1 and MCP-1 in the dose group was significantly lower than that in the light-brown and brown group,and the expression of TGF-?1 and MC P-1 The expression of TGF-?1 and MCP-1 in the high dose group was significantly higher than that in the low,middle and high dose groups.Conclusion 1.Kaijie Decoction can reduce the content of BUN,Scr and P in CRF model rats,protect residual renal unit function and delay the process of chronic renal failure.2.Kaijie Decoction by reducing the IL-6,TNF-? content to improve the CRF "micro-inflammatory state" to protect renal function.3.Kaijie Decoction by inhibiting the expression of TGF-?1,MCP-1 in renal tissue,delay the process of CRF fibrosis.
