The Influence Of Huangqibaoxin Decoction On Ventricular Remodeling And Its Methanism Research In Rats With Dilated Cardiomyopathy

Objective:To observe the influence of Huangqibaoxin(HQBX) Decoction on heart functions and ventricular remodeling in rats with dilated cardiomyopathy(DCM),through the observation and measurements of Echocardiography,Neuroendocrine and cardiac remodeling related indicators,with the addition of pathological morphology.Methods:Sixty rats were divided into the normal group (8 rats)and the rest were made to DCM model rats(52) which were given intraperitoneal injection of doxorubicin for eight weeks.Echocardiography was made to choose the DCM rats, then randomly divided them into four groups:Group HQBX (11) was given HQBX decoction 5ml/kg·d-1,group Captopril(ll) received captopril 50mg/kg·d-1,the last group adopted(11) a combination of HQBX decoction and captopril as the same volume mentioned. The rest of 11 rats were set to model group (NS,ig),and the normal group(8) had continued to been detected.Echocardiography was made again to explore heart functions including LVEDd,LVESd,EF and FS.Left ventricular mass index(LVMI)was also explored.ELISA(enzyme-linked immunosorbent assay) was used to assess the levels of AngⅡ、BNP、ST2,then using HE staining to see myocardium shape,and Picrosirius staining to differenciate collegen volume fraction(CVF). Meanwhile western blot and IHC were taken to semi-quantitatively analyse the levels of MMP-9 and TIMP-2.Results:Compared with the model group,treatment groups’LVEDd,LVESd,EF and FS change remarkably (P<0.05),AngⅡ、BNP and ST2 greatly reduced (P< 0.01),LVMI and CVF all notably decreased (P<0.01),the levels of MMP-9,TIMP-2 also have significant differences (P<0.01)Conclusions:HQBX decoction can improve heart function and relieve ventricular remodeling in rats with DCM,also it makes better effect together with captopril.
