Bamboo has the good biological activity of antioxidation, anti-aging, reducing blood pressure, regulating the immune system, anti-tumor and so on. Eucommia has the good biological activity of absolutely reducing blood pressure, tonifying kidney, analgesic, anti-inflammatory, improve immunity and anti-cancer activity. This paper, bamboo leaves extract (BLE for short) and Eucommia ulmoides extract (EUE for short) was studied. Firstly, the high performance liquid chromatography (HPLC) method was used to determine their composition and content. Then MTT method was used to measure the inhibition of their single component on the proliferation of human colorectal cancer cells (HCT116), cervical cancer cells (Hela) and human umbilical vein endothelial cells (HUVEC). Furtherly, screened the single component that both anticancer and anti-angiogenic activity, and simply studying its mechanism of action, in order to develop anti-angiogenic and anti -angiogenic drugs, main results are as follows:1.The content of Orientin Homoorientin Vitexin Isovitexin Caffeic acid and Chlorogenic acid in BLE were 4.47%,5.26%,0.37%,1.14%,0.07% and 0.42% respectively; the content of Geniposidic acid Chlorogenic acid in EUE were 13.29% and 3.41% respectively.2.The method of MTT and the human colorectal cancer cell line HCT116 were used to Study the anticancer activity of the seven components that mentioned above and the positive controlling drug. The results showed that the inhibition effect of isovitexin on proliferation of HCT116 cells was best.The method of MTT and Hela were used to Study the anticancer activity of seven kinds of single ingredient and positive control drug. The results showed that the inhibitory effect of the seven single components on the proliferation of Hela cells was not better than the positive controlling drug, but Vitexin Isovitexin and Chlorogenic acid is relatively good, When the mass concentration of chlorogenic acid increases from 40 μg/mL to 640 μg/mL, The inhibition rate increases from 0.41% to 40.67%, But Vitexin and isovitexin presented different law.3. The method of MTT and human umbilical vein endothelial cells HUVEC was used to study the anti angiogenic activity of seven kinds of single and positive control drugs. The results showed that the inhibitory effect of the seven single components on the proliferation of HUVEC cells was not better than the positive controlling drug. but, Vitexin Isovitexin and Chlorogenic acid was relatively good. Beside, this paper also received the changes of their relationship of dose-effect.4. The methord of Annexin V and PI and flow cytometry was used to study the effect of BLE and Isovitexin on the apoptosis of HCT116 cells.The results were as follows:BLE has a certain effect on the apoptosis of HCT116 cells. But with the increasing of its concentration, the induction effect was gradually reduced. EUE has a certain inducing effect on the apoptosis of HCT116 cells, but the concentration dependence was not obvious. Isovitexin had a certain inductionon on HCT116 cells apoptosis, but with the increasing of the concentration, inducing effect gradually decreases. when the concentration were 80 μg/mL,160 μg/mL,320 μg/mL, Correspondingly. the apoptosis rate of HCT116 cells were 12.30%,9.00% and 9.20%, respectively. Which were higher than the processed group of BLE and the treatment group of EUE at 200 μg/mL,400 μg/mL and 800 μg/mL respectively. It was showed that isovitexin had a better inducing effect on apoptosis of HCT116 cells,which lays a certain foundation for further study of anti-tumor mechanism.
