ObjectiveTo explore the relationship between IgA nephropathy with Damp-heat body constitution and the gene polymorphism of IL-10-1082/-819/-592 and ICAM-1 K469E/G241R. To find the genetic susceptibility of IgA nephropathy and Damp-heat body constitution, so to service for the clinical diagnosis better.MethodCollect the volunteers from in-patient department and Health Check Centre in Guangdong hospital of Chinese medcine. All the participants were assessed by Chinese body constitution and those who up to the standard will be included in the study. We set four groups:IgA nephropathy with Damp-heat body constitution, IgA nephropathy with Balanced body constitution, the health with Damp-heat body constitution and the health with Balanced body constitution.3ml venous blood was collected to extract DNA. Then the DNA was expanded by PCR, cheack the gene section by electrophoresis to detect whether the section is the purpose gene. Finally sent the products to biological company to sequence. All the genotypes were judged by the chromas 2 software. Then use SPSS19.0 software to statistical analysis, find out the different genotypes among each groups.Result1.98 people were included in this study and 29 cases were IgA nephropathy with Damp-heat body constitution,21 cases were IgA nephropathy with Balanced body constitution,22 cases were health with Damp-heat body constitution,26 cases were health with Balanced body constitution. There was no no statistical difference in age and gender among each groups.2. To test the Hardy-Weinberg law of the five loci gene frequency above, we find that the gene frequency of IL-10-1082/-891/-592 conformed to the law and it means that it has a good group representative (p>0.05), but the gene frequency of ICAM-1 G241R/K469E was not(p<0.05).3. To analyze the genotypes and alleles of the 5 sites above, only IL 10-1082 site has differences among the groups(p<0.05). The IL-10-1082 was mainly composed of AA genotype(87.8%), GG genotype was not detected, and Alleles are mainly composed of A(93.9%).4. Further study of IL-10-1082 loci among groups, we found that AA genotype and A alleles were higer in IgA nephropathy than the health(94%:81.3%and 97%:90.6%). In people with Damp-heat body constitution, the AA genotype were higher in people with IgA nephropathy than health people. But in health people, the GG genotype and G alleles were higher in those with Damp-heat body constitution than those with Balanced constitution.Conclusion1.The AA genotype and A allele of IL-10-1082 loci may be a genetic susceptibility factors of IgA nephropathy. The one with such genotypes or allele maybe more prone to IgA nephropathy.2. In people with Deap-heat body constitution, the AA genotype will higher in those with IgA nephropathy than the health people. But this is not in balanced body constitution. Maybe this means that there exists a correlation between IgA nephropathy and Damp-heat body constitution. So Damp-heat body constitution people may be more susceptible to IgA nephropathy。3. In the health people, the GG genotype and G alleles will higher in those with deap-heat body constitution than that with balabced constitution, but this is not in IgA nephropathy. And IgA nephropathy is always related with the AA genotype and A alleles.
