Screening Of Differentially Expressed Genes And Their Functions In Damp-heat Constitution Of The Chinese Medicine

ObjectiveThe purpose of our study was to investigate whether the damp-heat constitution had some difference at the genetic level comparing with other types of constitutions in TCM, so as to understand the nature of damp-heat constitution at the micro level and their relationship with disease.MethodsAccording the 9 kinds of Chinese medicine constitution classification and decision criteria, proposed by Wang Qi,we collected 8 cases of damp-heat constitution and 8 cases of peaceful people, which had pass general physical examination and biochemical indicators. And then each one would be collected fasting blood 3ml, that used to extract total RNA in 4 hours by RNAprep pure Blood kit. The total RNA was Stored in -80℃,it would be send to RiboBio Biological Technology in 1 week to finish Gene expression seq, in order to screen the genes with significant differences in damp-heat constitution group (q-vallue＜ 0.001 and| log2 (a Fold change)|＞ 1) and analyze their main functions.ResultsIn our study, a total of 27 significant different genes related to damp-heat constitution has been found,18 of them are raising genes, and 9 are down-regulated genes. Most of them mainly on the immune system, autoimmune diseases such as type I diabetes, autoimmune thyroid disease, systemic lupus erythematosus (sle), tumor. Some of them related to steroid metabolism, folic acid metabolism, extracellular matrix cracking, code binding protein or nucleosome structure protein, etc. There are 5 signaling pathways of enrichment. One is about phagosome, two of them on immune defense, another two are respectively associated with type Ⅰ diabetes and autoimmune thyroid diseases.ConclusionAccording to our research, damp-heat constitution people really have their own specific genes, which are obvious different from peace-constitutions, particularly in aspects of immune defenses. As a result, damp-heat constitutions may have a ill tendency on prompt, type I diabetes and AITD disease etc, which caused by autoimmune disorders.What’s more, it is closely related to the whole process and prognosis of inflammatory disease in kidney. In addition, this type of constitution aslo perfoms a disorder on cholesterol metabolism, so they more likely to have metabolic diseases. Therefore, it is very important to correct the damp-heat constitution as soon as possible. And it is still need more large samples to study the nature of the physical characteristics deeper, and its relationship with other diseases.
