Hyperlipidemia is a major cause of atherosclerosis and cardiovascular related diseases, so the development of hypolipemic active ingredients becomes a hot research in recent years. As a kind of physiologically active substances, Auricularia auricular polysaccharides has been paid increasing attention recently.In this study, Auricularia auricular was treated as aresearch object, its lipid-lowering active ingredient in vitro were screened, and hypolipemic activity was evaluated in vivo by administration to analyze its effect on the prevention and treatment of hyperlipidemia, promote the development and utilization of Auricularia auricular polysaccharides. The specific results are as follows:1.Response surface optimizes the process parameters of Auricularia auricular polysaccharides by high pressure homogenization, the results shows:homogeneous time:24.91min, homogenizing pressure:24.66MPa, solid-liquid ratio:1:76.15, the theoretical value of Auricularia auricular polysaccharides was up to 40.74%.2.Under the optimum parameters conditions,0.1MHCl, distilled water, 0.1MNaOH as solvent extraction to extract three kinds of Auricularia auricular polysaccharides, The sodium glycocholate and sodium taurocholate serve as the evaluation index, The results showed that the ability of alkaline Auricularia auricular polysaccharides adsorbed sodium glycocholate and sodium taurocholate was best, acid extraction was second, water extraction is the lowest;Alkali extraction graded by ethanol precipitation, getting 6 section alkaline extractio is 50%,60%, 70%,80%,90%,100% respectively. The capacity binding bile salts in vitro was analysised. The results showed that:The adsorption rate of 70% alcohol precipitation alkaline extraction (J-70) to sodium glycocholate and sodium taurocholate are superior to other fractions, Therefore, it was elected as raw materials for animal experiments.3.Biulding mice hyperlipidemia model by feeding high-fat diet at the same time feeding J-70. To investigate the effect of J-70 for preventing hyperlipidemia. The results showed that:J-70 for long-term consumption of high-fat diet mice had a significant hypolipemic effect. J-70 can slow the weight gain of mice, and reduce liver index and atherosclerosis index, Wherein the effect, the high dose group was significantly;The results of lipid horizontal displays:J-70 can increase high-density lipoprotein cholesterol (HDL-C), lower cholesterol (TC) and triglyceride (TG) content, Wherein the groups,the middle dose group was significantly,the high dose group is best. Low-density cholesterol (LDL-L) content of J-70groups is decreased, But the effect was not significant; The measurement results of lipid metabolism related enzymes showed J-70 can Increase the content of lipid metabolism-related enzyme acyltransferase (LCAT) and increase the role of lipoprotein lipase (LPL) and hepatic esterase (HL) activity,and the high-dose group is significant; in terms of antioxidant, J-70 groups could significantly increase the activity of total superoxide dismutase (T-SOD) and glutathione reductase (GSH-PX),It has a certain antioxidant effect.4.Established hyperlipidemia mice model by feeding high fat diet method,After the model is built,J-70 were gavage for treatment, the hypolipemic effect of J-70 of high-fat mice were evaluated. The results showed that:J-70 for controlling weight gain had a significant effect, the effect of high-dose group was significantly, while J-70 can inhibit the rise in the index of liver, and increase the spleen index of mice, thus enhancing the immune function; Atherosclerosis index (AI) showed J-70 groups were able to significantly reducen the AI value of hyperlipidemia mice, it indicatdes that J-70 can hinder the generation of atherosclerosis; The results of lipid horizontal displays, J-70 can reduce the levels of TC, TG and LDL-C, the middle dose group was significantly and the high dose group the most significant, HDL-C content in terms of the test substance in each group increased,They showed highly significant, it indicates that J-70 is able to control the blood lipid levels of hyperlipidemia mice;The 1 measurement results of ipid metabolism related enzymes show:J-70 groups could significantly increase LCAT content, the activity of LPL and HL,and in a dose-dependent;In the antioxidant test, J-70 can be significantly increased T-SOD and GSH-PX activity, and has a effective treatment on excessive oxidative damage caused by hyperlipidemia.In this research, the response surface method was used to process the optimization of high-pressure homogenizer for extracting Auricularia auricular polysaccharides, By the test in vitro screens the optimal hypolipemic fragment,it was J-70 (70% alcohol precipitation alkali solution under high pressure homogenizer optimization parameter),and its composition was analyzed, its preventive and therapeutic effects were analyzed and compared, The results showed that:J-70 has the potential hypolipemic function.
