Nepeta angustifolia C. Y. Wu pertains to the Nepeta genus of Labiatae family.It grows in hillside,roadside,the lawn and the edge of forest,at the elevations between 3600 and 4600 m.It’s mainly distributed in the central and south areas of Tibet.Mainly produced in Tibet Lhasa,gyantse, etc.The Nepeta angustifolia C.Y.Wu is the unique medicinal materials for Tibet.It’s flowers and leaves is used medicine in Tibet folk. Nepeta angustifolia C. Y. Wu plays the roles of causing resuscitation and restoring consciousness,anti-inflammation and analgesic,In the Tibetan medicine is mainly used to treat dizziness and apoplexia,pain,cerebral hemorrhage,traumatisme and pestilence,etc.Now there had less study about Nepeta angustifolia C. Y. Wu,its standard contains in the ?Tibet autonomous region local medicinal quality standard? only its properties,processing,sexual flavour and functional attending are expounded.The study of Nepeta angustifolia C. Y. Wu about its pharmacological activity and chemical composition almost in a blank state,and its clinical application is confined to the traditional prescription.All these seriously hinder the further development of Nepeta angustifolia C. Y. Wu.For systematic study Nepeta angustifolia C. Y. Wu chemical constituents and pharmacological activities.In this thesis,the antioxidant capacity of Nepeta angustifolia C. Y. Wu different polar parts was studied, its chemical composition was extracted,separated and structural identification,the anti-inflammatory and abirritation pharmacological activities were studied.The results are as follows:(1)The Nepeta angustifolia C. Y. Wu whole plant was extracted with 70%ethanol.The exteract was extracted by petroleum ether,dichloromethane,ethyl acetate,n-butyl alcohol, ethanol.DPPH scavenging system,prussian blue method and H2O2/Fe2+ salicylic acid detection system were used to measure the antioxidant activity,scavenging activity and reducing power.The antioxidant activity of extract was compared with that of vitamin C and rutin.Three oxidation experiments show thatNepeta angustifolia C. Y. Wu extract by petroleum ether,dichloromethane, ethyl acetate,n-butyl alcohol,ethanol extracts had different levels of antioxidant effect.The antioxidant activity of extractive and samples concentration showed a dose-response relationship.In H2O2/Fe2+ salicylic acid detection system to test scavenging activity when dichloromethane,ethyl acetate,ethanol, n-butyl alcohol and petroleum ether extract concentration reached in 1.0g.L-1 the antioxidation were greater than 60%,and the concentration of ethyl acetate reached in 0.7g.L-1 the antioxidation even higher than the positive control of rutin and Vc.For Nepeta angustifolia C. Y. Wu as a natural antioxidant and free radical scavenger provides a theoretical basis.(2)In this experiment,it was studied that anti-inflammation and analgesic effect of extracts from different solvents extraction layer of Nepeta angustifolia C. Y.Wu.The mouse were randomly divided into seven groups, as the control(treated with normal saline),positive control(with prednisone or aspirin),extracts from different solvents extraction layer of Nepeta angustifolia C. Y. Wu( groups treated with water layer,n-butyl alcohol layer,ethyl acetate layer,methylene chloride layer,petroleum ether layer).Models of mouse ear edema induced by xylene and peritonitis induced by xylene were used to reasearch the anti-inflammatory effects,the acetic acid caused writhing and hot-plate test in mouse were taken to reasearch the analgesic effects.The results showed that the mouses’ ear swelling rate,OD value,writhing times,the pain thresholds were obviously lower in the treatment group and the positive control group than the control group.The conclusion is that extracts from different solvents extraction layer of Nepeta Angustifolia C. Y. Wu possess anti-inflammation and analgesia capacity, especially ethyl acetate layer, methylene chloride lay, petroleum ether layer.(3)Nepeta angustifolia C. Y. Wu whole plant after remove impurities all18 kg, after shear to infiltration percolation barrels,with 70% ethanol and material ratio of 1:20(kg/L) percolation extract for 15 days,diacolation liquid was concentrated under vacuum to 1:2(kg/L),with petroleum ether,ethyl acetate and water saturated n-butanol to extract and separate in turn.Final,acquired petroleum ether,ethyl acetate,water saturated n-butanol and water extraction layer extraction respectively220 g,400g,750 g,1500g.The ethyl acetate layer by silica gel column chromatography,gel column chromatography,thin-layer chromatography, ODS column and preparative liquid phase and other separation methods to separate, and the compounds isolated were identified by nuclear magnetic chromatography,11 compounds were isolated fromthe Nepeta angustifolia C.Y.Wu,respectively,oleanolic acid,betulinic acid,ursolic acid,β-sitosterol,hesperidin,angelicin, bakuchiol, methyl rosmarinate,blumenol C glucoside, psoralen,rosmarinic acid.(4)The powder of different medicinal parts of Nepeta angustifolia C. Y. Wu and Schizonepeta tenuifalia Briq.were sealed in the headspace bottle,and we aimed to examine volatile components of different medicinal parts(flower,stem,leaf and whole plant) of Nepeta angustifolia C. Y. Wu and Schizonepeta tenuifalia Briq stem.by headspace gas chromatography-mass spectrometry(HS-GC/MS).The volatile constituents were identified by retrieving the NIST library, the relative percentage contents of which were calculated with area normalization method to examine their difference.It turned out that the main volatile component from Nepeta angustifolia C. Y.Wu was nepetalactone(73.24% in stem). Among different medicinal parts of Nepeta angustifolia C. Y. Wu 21 volatile constituents were characterized in leaves,17 compounds were identified in whole plants.While,the volatile components of Schizonepeta tenuifalia Briq. stem mainly contained menthone,pulegone and cinene,18 constituents were identified in which.Our results showed significant difference about volatile components and provided the important information to explain the difference of efficacy and cured diseases between Nepeta angustifolia C. Y. Wu and Schizonepeta tenuifalia Briq.
