Traditional Chinese medicine(TCM)is an analytical system with too much complexity.Usually,there are a lot of chemical components in single herbs and most of them are with low concentration.As a result, commonly analytical instruments and detecting approaches can not make it possible to qualitatively and quantitatively determine chemical components from single herbs completely.With the advent of hyphenated chromatographic instruments,such as GC-MS and HPLC-DAD,and the rapid development of some chemometric multi-resolution techniques like HELP,it is possible to completely conduct the qualitative and quantitative analysis for TCM by use of the combined information on separation and identification produced by chromatography and spectrometry.In this study,three herbal pairs(HPs),Rhizoma Coptidis-Radix Glycyrrhizae,Herba Schizonepetae-Ramulus Cinnamomi and Zingiber officinale Rosc.-Ramulus Cinnamomi,were investigated with two-dimensional hyphenated chromatographic instruments and chemometric resolution methods.The main contents of the thesis are as follows:1.Part one(chapter 2 and chapter 3).Qualitative and quantitative analysis of components in the decoctions and precipitation of herbal pair Rhizoma Coptidis-Radix Glycyrrhiza with HPLC-DAD method.In chaPter 2,the content changes of berberine and glycyrrhizic acid were analysized in the decoctions of the single herb and herb pairs.With the help of chemometric resolution approaches,such as heuristic evolving latent projections(HELP),upon the two-dimensional data produced,pure chromatogram and UV spectra of each component can be obtained.With the combined information on retention times and UV spectra,chemical components in decoctions of the single herb and herbal pairs were compared qualitatively and quantitatively.The conclusion was drew that the contents of berberine and glycyrrhizic acid in the decoctions of HPs were lower than in the single herb.In addition,most components of Rhizoma Coptidis could be found in the HPs while only half components of Radix Glycyrrhiza could be found.With the same methods,in chapter 3,chemical components of precipitation which could dissolve in the simulated gastric fluid and intestinal fluid were compared qualitatively and quantitatively.The result showed that the solubility of precipitation in the simulated intestinal fluid was better than simulated gastric fluid,the primary components in both simulated gastric fluid and intestinal fluid was berberine.2.Part two(chapter 4).The method of water-steam distillation was used to extract the essential oils from single herb Herba Sehizonepetae, Ramulus Cinnamomi,Zingiber offieinale Rose.,and herbal pairs Herba Sehizonepetae-Ramulus Cinnamomi,Zingiber offieinale Rosc.-Ramulus Cinnamomi.Chemical components of these essential oils were analyzed by gas chromatography-mass spectrometry(GC-MS)together with HELP method.The common compounds between the single herbs and HPs were identified by the method of altemative moving windows factor analysis (AMWFA).The result showed that 12 chemical compounds of HPs Herba Schizonepetae-Ramulus Cinnamomi were from Herba Sehizonepetae,16 chemical compounds were form Ramulus Cinnamomi,6 new chemical components were only in HPs,there are 14 chemical compounds among the three systems;Another HPs Zingiber offieinale Rosc.-Ramulus Cinnamomi,25 chemical compounds of HPs were from Zingiber officinale Rosc.,21 chemical compounds were form Ramulus Cinnamomi, 3 new chemical components were only in HPs,there are 10 chemical compounds among the three systems.The results of the study will lay the foundation for the further research of the theory of compatibility of TCM and clinical work.
