Objective: Cirrhosis is a variety of end-stage liver stage development of inflammatory diseases, clinical diagnosis should not only consider the case of a single disease, the need for a comprehensive assessment of the general condition, cirrhosis of the liver disease occurs, it may be accompanied by other complications, cirrhosis induced other parts related diseases are a leading cause of cirrhosis mortality. From the point of view of cirrhosis pathology analysis, liver tissue surface view has undergone significant change, active liver cells was not observed under the microscope, View of a large number of dead cells filled, typical structural changes in liver cirrhosis. The development and progression of cirrhosis complicated, note that the occurrence of a variety of health, for example, abdominal effusion, acute bleeding and jaundice, advanced liver function tests showed significant abnormalities, disease continue to progress, the final result of liver failure in the end. Currently, the clinical diagnosis of cirrhosis is no good cure effect, and the selectivity of drugs, drug abuse treatment will increase the burden on the liver, delay treatment, we need to find new treatments to improve the treatment of liver cirrhosis. the main source of bone marrow mesenchymal stem cells to repair damaged when as hepatocytes, functional liver tissue can be improved, and compared with other means of treatment of liver cirrhosis, bone marrow mesenchymal stem cells transplantation easily obtained, no rejection, safety and relatively inexpensive, etc. This experiment used the bone marrow of neonatal rat to culture bone marrow mesenchymal stem cells’ primary cell, based approach to carbon tetrachloride, on experimental mice with model establishment of liver cirrhosis, then, transplantation of its value-added ability of bone marrow mesenchymal stem cells, and take effective means to detect the degree of recovery of liver cirrhosis, analysis of the final result. Animal experiments and basic research provide more conclusive evidence and strong support for further clinical application.Methods: The anatomy of male suckling mice, the extraction of bone marrow, bone marrow adherent culture method for bone marrow mesenchymal stem cells, primary cells were digested,get bone marrow mesenchymal stem cells within three generations; in cultured cells surface labeled monoclonal antibody CD11 b, CD29, CD31, CD14, CD34, CD45, CD73, CD90, CD105, it was detected by flow cytometry; use of carbon tetrachloride-based, supplemented by a method ofphenobarbital, for the BALB/C male mice modeling cirrhosis, normal food and water for the BALB/C male mice were used as the control group, preparation of pathology, both results were cf.Reference; migration path can be used in two ways: the approacho on hepatic artery and tail vein,after the successful transplantation, observed in the experimental group of mice over time, liver tissue was visually observed, analysis of pathology report results, it gets the results of treatment of cirrhosis of data.Results: Get within three generations, value-added capability and a more purified bone marrow mesenchymal stem cells; carbon tetrachloride-based approach to BALB/C mice were male modeling cirrhosis, pathology results showed successful cirrhosis model; tail vein and hepatic artery cirrhosis mice transplanted bone marrow mesenchymal stem cells, the pathology report showed the degree of cirrhosis of the liver have been well improved to some extent, effects of the two ways to treat fairly.Conclusion: Flow cytometry results illustrate that positive expression rate of CD105, CD73,CD29, CD90 were greater than or equal 95%, strong positive expression in cultured cells surface were showed, that positive expression rate of CD14, CD11 b, CD34, CD45, CD31 were Less than or equal 2%, negative expression were showed. It shows the cells cultured in line with the basic characteristics of bone marrow mesenchymal stem cells, by whole bone marrow adherent culture method for culturing bone marrow mesenchymal stem cells, Primary cells were digested and passaged within three generations and obtain cultured cells, tne ability to add value is formidable,cell types were purified, it can be transplanted for the treatment of disease. Methods based on the use of carbon tetrachloride cirrhosis model mice were established, liver cirrhosis change significantly, histology results are also consistent with, it is a description of this method can be carried out on mice with liver cirrhosis modeling. Hepatic artery and tail vein transplanted BMSCs certain number of cells transplanted into mice with liver cirrhosis, liver tissue was visually observed, get on HE staining and pathological tissue sections, the results showed that liver cirrhosis improved to a certain extent, two ways have no significant differences in the effect of liver cirrhosis.
