Objective : To assess the differences between the therapeutic effects of two different transplantation approaches of bone marrow mesenchymal stem cells(BMSCs)on mice model of cirrhosis.Methods : The cirrhotic model was constructed by subcutaneous injection of mixture of CCL4 olive oil.Taking 36 cirrhotic mice divided into 3 groups randomly:no transplantation group(n=12)?tail vein transplantation group(n=12)?spleen transplantation group(n=12);BMSCs were isolated and cultured by the method of whole bone marrow adherent,and choose the passage3 to analyze by flow cytometry.We selected tail vein and spleen injecting approaches to graft BMSCs.Four weeks later,liver function tests was determined,and liver fibrosis was detected by HE stain,Masson stain and ?-smooth muscles actin(?-SMA)immunohistochemical analysis.Result:liver function?histopathological examination and immunohistochemical analysis were ameliorate significantly after transplantation,two injecting approaches contrasted to control group showed a statistical differences(P<0.05);while the comparison between the two ways did not show statistical differences markedly(P>0.05).Conclusion : Injecting BMSCs via tail vein or spleen approaches have the same curative effect on the liver function and cirrhotic degree in mice cirrhosis model.
