[Objective]To detect the expression of epithelial-mesenchymal transition biomarkers and MDM2 in non small cell lung cancer tissues, and evaluate the clinical significance of it.[Methods]The quantitative RT-PCR and immunohistochemistry were used respectively to detect the mRNA and protein expressions of E-cadherin, N-cadherin, Vimentin and MDM2 in NSCLC tissues and cancer adjacent normal tissues. The SPSS 17.0 statistic software was used to analyze the correlation between the mRNA or protein expressions and the clinic-pathological agents of the patients with NSCLC.[Results]Quantitative RT-PCR results showed that Compared with adjacent normal tissues, N-cadherin, vimentin and MDM2 mRNA were overexpressed in NSCLC tissues(P<0.05), E-cadherin mRNA was downexpressed in NSCLC tissues(P<0.05). In non small cell lung cancer, the expression of N-cadherin, vimentin and MDM2 mRNA were significantly correlated, while E-cadherin mRNA expression was negatively correlated with MDM2 mRNA expression.E-cadherin mRNA expression is associated with gender, tumor size and the degree of differentiation, and patient age,smoking status,histological type and lymph node metastasis has no significant relation. N-cadherin and vimentin mRNA expressions are associated with tumor size, the degree of differentiation, and gender,patient age,smoking status, histological type and lymph node metastasis has no significant relation. MDM2 mRNA expression is associated with tumor size, the degree of differentiation and lymph node metastasis,and patient age, smoking status and histological type has no significant relation.Immunohistochemisty results showed that Compared with adjacent normal tissues, N-cadherin, vimentin and MDM2 protein were overexpressed in NSCLC tissues(P<0.05),E-cadherin protein was downexpressed in NSCLC tissues(P<0.05).In non small cell lung cancer, the expression of N-cadherin, vimentin and MDM2 were significantly correlated, while E-cadherin expression was negatively correlated with MDM2 expression.E-cadherin expression is associated with gender, tumor size and the degree of differentiation, and patient age, smoking status,histological type and lymph node metastasis has no significant relation. N-cadherin and vimentin expressions are associated with tumor size, the degree of differentiation, and gender,patient age, smoking status,histological type and lymph node metastasis has no significant relation.MDM2 expression is associated with tumor size, the degree of differentiation and lymph node metastasis, and patient age, smoking status and histological type has no significant relation.[Conclusion]In non small cell lung cancer, he over expression of MDM2 may promote tumor epithelial-mesenchymal transition, the combined detection of MDM2 and epithelial-mesenchymal transition related molecular markers by quantitative RT-PCR and immunohistochemistry, can be used as important reference to judge the malignant degree, metastasis potentiality and prognosis of non small cell lung cancer.
