| Objective To investigate the expression and role of human Dachshund homolog 1(DACH1) in the tongue squamous cell carcinoma(TSCC).Methods Fifty-one patiens’ tumor specimens with TSCC,who had surgical removal of the tumor with concurrent neck lymph dissection,were collected in the Affiliated Hospital of Qingdao University from 2011 to 2013. All patients had no radiotherapy,chemotherapy or intervention therapy before operation.All the patients were calssified on the basis of age,gender,clinical stage,gross type,differentiation,lymph node metastasis.Immunohistochemistry were used to explore the expression of DACH1 in 51 samples of TSCC and paired adjacent tissues accompanied by analyzing its expression relationship with clinical pathological indexes.Real-time polymerase chain reaction and Western blot were used to detect the expression of DACH1 in nine samples of fresh tumor and adjacent tissues.Results The results showed that about 70.6%(36/51) of squamous cell carcinoma was lower than adjacent tissues in terms of expression level of DACH1.Moreover,there was significance differences between that(P<0.05),and such lower expression was associated with poor differentiation of tumors, late clinical stage, and lymph node metastasis(P<0.05).Moreover,Real time RT-PCR and Western Blot test results showed that DACH1 m RNA and protein expressed in tongue squamous carcinoma tissues were lower than that in the adjacent tissues(P<0.05).Conclusion Expression level of DACH1 is lower in tumors than in adjacent tissues,which have relationship with poor differentiation of tumors, late clinical stage and lymph node metastasis.DACH1 may provide an effective targeted drug therapy sites for TSCC patients.Objective To explore the role of human Dachshund homolog 1(DACH1) on the proliferation,apoptosis,adhesion,invasion and metastasis of SCC-25 cells.Methods Western Blot detected the respectively expression of DACH1 in CAL-27,Tca 8113 and SCC-25 cells.Lentiviral overexpression vectors harboring the DACH1 gene were constructed for the transfection of SCC-25 cells as experimental group.DACH1 empty vector plasmid,as negative control,and its packaging plasmid which was blank control were extracted and cotransfected into SCC-25 cells.Effect of overexpression DACH1 on cell proliferation determined by MTT and colony-formation assay.Transwell and adhesion assay tested the role of DACH1 over expression in adhesion,invasion and metastasis ability.Flow cytometry detected the role of DACH1 over expression in apoptosis.Results Expression level of DACH1 in SCC-25 is the significant lowest than that in CAL-27 and Tca 8113(P<0.05).Overexpression of DACH1 significantly inhibits the ability of proliferation and ability of colony-formation,as well as the process of invasion and metastasis, while promotes apoptosis in SCC-25 cells(P<0.05).Conclusion Overexpression of DACH1 may closely related to the progress in tongue squamous cell carcinoma,which significantly inhibits ability of proliferation,adhesion,invasion and metastasis in SCC-25 cells and promote its apoptosis.Therefore,overexpression of DACH1 in TSCC have ability in restraining malignant development.It’s hopeful to provide new clinical treatment for TSCC. |
