Object1.To investigate changes in brain-derived neurotrophic factor(BDNF) in the hippocampus of rats with post-traumatic stress, and the significance of such changes.2. To observe the expression and significance of BDNF m RNA in the blood of rats with Post-traumatic stress disorder. And mechanism of Paroxetine treatment PTSD。Methods1. To investigate the effect of traumatic stress on the hippocampal BDNF expression research process:choose 6-7 periodontal health clean male wistar rats,A total of 32。3 model groups(1d, 7d, and 14 d after model construction) plus a normal control group were set。To establish PTSD model 。 Detect the expression of BDNF levels in hippocampal cells,Compared to differences between normal group and stress groups of BDNF expression in hippocampus by enzyme-linked immunosorbent assay.2. Discussion The expression levels of rat blood BDNFm RNA of PTSD(Post Traumatic Stress Disorder, PTSD).research process: choose 6-7 periodontal health clean male wistar rats,A total of 32, and model groups, treatment group(paroxetine) plus a normal control group were set 。 To establish of PTSD model, After the model is establish successful, the treatment group(paroxetine) in accordance with the 10 mg /(kg · d) dose given paroxetine for 14 days.After14 days, to detect changes of BDNF m RNA in the blood of three groups rats by the real-time quantitative PCR(q RT-PCR)3. Statistical analysis: The Shanghai Hui Cheng Psychology Test Software was used to transform original data.Data was analysed by SPSS17.0 software. the nonparametric tests U Mann-Whitney test was used to analyse the non-normal data.Spearson correlation analysis was applied to perform correlation analysis. All tests were two-sided test, the test level is P<0.05.Results1.(1) BDNF levels of model lower than normal group, and the difference is statistically significant(P<0.05).BDNF levels of the normal group and that of 1d 、7d are not difference;there are differences with the 14 d, the difference was statistically significant.Change of BDNF levels is rather slow.2.(1) The level of BDNFm RNA expression was significantly higher than normal group, and the difference is statistically significant(P<0.05). In the treatment group rats,the level of BDNF m RNA in blood is higher than normal group, relatively low stress group, there was no statistically significant difference.Conclusions1. BDNF level decreases in rats suffering from post-traumatic stress, but such change is rather slow. BDNF is a potential marker for post-traumatic stress.2. The level of BDNF m RNA in blood of rat suffering PTSD is higher than normal group,BDNF m RNA may as a protective factor involved in the occurrence and developing process of PTSD, the impact of paroxetine on BDNF m RNA levels of PTSD Rats for further study.
