Objective: Cerebral infarction death from stroke on 4600000 people worldwide each year, and every year 1600000 people die of the disease in our country. Now its incidence shows the tendency of increase year by year.It has a high morbidity、low treatment and poor prognosis. The disease has serious harm to people’s health.This experiment by observing the combination of acupuncture with medicine on rat model which were used purgation and modified thread-occlusion method. The experiment indicators is EPCs number changes in different time points and VEGF in the rats’ hippocampus to explore the promotion and mechanism of vascular repair after hurt; Serum REDOX enzyme were studied to know the effect on the MCAO rats’ antioxidant capacity of Chinese medicine and acupuncture- medicine.Methods: 1. Selected 50 male SD rats. Balance raised all rats in 3 days, then divided them randomly, There were normal group(NG),control group(CG), MCAO group(MG), rhubarb + MCAO group(RMG). CG and RMG were feeded rhubarb for 7 days. MG and RMG were used method of modified thread-occlusion to manufacture ischemia reperfusion model rats. Indicators were the general condition, hemorrheology and EPCs. 2. Selected 40 male SD rats.Balance raised all rats in 3 days, then randomly divided them into normal group(NG), control group(CG), model group(MG), drug group(DG) and acupuncture- medicine group(AMG).Randomly selected 6 rats for NG.Give the rest of the 34 rats rhubarb for a week,. Manufacturing model and then using optical microscope to observe HE staining in rat hippocampus. 3. Detected each group rats serum REDOX enzyme. 4. Tested rats’ hippocampus VEGF protein with immunohistochemical method. 5. Tested rats’ EPCs in different time points.Results: 1. The model was successfully established. 2. HE result: hippocampal cells of MG layer decreases, clutter is not tight,uneven dyeing, the outline is not clear, see more nuclear pyknosis,unclear nucleolus, disappeared cytoplasm; hippocampal cells of DG were more closely rules, there were three or four layers of cells layer, visible nucleus pycnosis, cell staining is uniform, outline is clear; hippocampal cells of AMG rules with three or four layers of cells layer, rare nucleus pycnosis, cell staining is uniform, outline is clear. 3. Serum REDOX enzyme result: SOD of MG decreased, MDA, LDH and i NOS increases, SOD of DG compared with MG increased, MDA, LDH and i NOS decreased.SOD of AMG increased significantly compared with DG, and other three activity decreased significantly(P < 0.05). 4. VEGF result: VEGF positive cells of MG was increased.Compared with MG, VEGF positive cells of DG was increased(P < 0.05), while AMG was higher than DG(P < 0.05). 5. EPCs result: EPCs number of MG reduced and control group, DG increased compared with MG(P < 0.05), and compared with those two, AMG’s EPCs number increased.Conclusions: 1. The model was successfully established with method of purgation and modified thread-occlusion. 2. Through the results, we can see that Chinese medicine can reduce oxidase and increase VEGF and EPCs numbers.But the acupuncture- medicine treatment shows better effect. This may because of acupuncture- medicine treatment has other targets or pathways, but this conclusion needs to be studied further more.
