AIM:In order to certify aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li has the ability of antioxidation and analyze its mechanisms, We detected MDA,ROS,SOD,T-AOC in heart,brain,kidney and blood serum of mice induced by D-galactose, together with macro-and micro-scopical changes were examined at the same time .METHODS:108 Kunming mice were divided into six groups at random, which were control group (which was abbreviated to"CG group"in the following), model group (which was abbreviated to"MG group"in the following), Salvianolic acid B control group (which was abbreviated to"SB group"in the following), high dose of Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li group (which was abbreviated to"HS group"in the following), middle dose of Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li group (which was abbreviated to"MS group"in the following), low dose of Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li group (which was abbreviated to"LS group"in the following). There were 18 mice in each group. We duplicated the model of oxidative stress mice with subcutaneous injection of 5% D-gal at the dose of 0.1 ml?10g-1?d-1 except CG group which was injected with the same volume of saline solution. And then 6 group were continuously filled medicine or saline through the stomach simultaneously CG and MG group were filled with saline and SB, HS, MS, LS group were given medicine. After 30 days , the MDA,ROS,SOD activity, T-AOC in heart,brain,kidney tissues and blood serum were detected. At the same time We observed morphologic changes of three tissues under light and electron microscope.RESULTS:1 Effects of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li on pathomorphology in brain,heart and kidney of OS miceThe pathomorphology under light mirror lens indicates that cerebral cortex of mice brain tissue and cardiac muscle fibers of mice heart tissue taking up the pathological changes of inflammation.But the cortex of kidney tissue does not take up any obvious changes. The pathomorphology under electron microscope indicates that the pericellular membrain,endocytoplasmic reticulum and nulear membrain are damaged or suffers fragmentation in cerebral cortex and cardiac muscle fibers. The pathological changes of tissues was serious in model group. The pathological changes of each medical group was lighter than that of model group.2 Effects of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li on MDA,ROS,SOD and T-AOC in brain,heart and kidney of OS miceThe contents of MDA,ROS rised and the activities of SOD,T-AOC decreased significantly in brain,heart,kidney and blood serum of MG mice( P<0.01 or P<0.05 ).The drug of each medical group could relieve the damage induced by D-gal( P<0.01 or P<0.05 ).CONCLUSION:The MDA,ROS contents of MG group comparing with that of CG group were raised significantly, and the activities of SOD,T-AOC was lower, which indicated that there were typical oxidational injury. Aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li is the effective medicine and has certain effect. According to the experiment aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li could increase the activities of SOD, T-AOC and reduce the contents of ROS, MDA, which demonstrated this medicine can remove free radical and increase the activities of antioxidase. In other way it can resist oxidative stress.
