ObjectiveTo evaluate the effects of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li on blood pressure, blood pressure variability and end organ damage in spontaneously hypertensive rats. Explore the mechanisms of the effects.Methodsthe experiment is divided into single-dose study and prolonged administration study.Part one16 rats were randomized into 2 groups: Salvia miltiorrhiza Bge f alba.C.Y.Wu. etH. W. Li group and normal sodium group. Normal Wistar-Kyoto (WKY) rats were used as negative control. Salvia miltiorrhiza Bge f alba.C.Y.Wu. et H. W. Li concentration was 2ml/kg (15 g.kg-1) by gastric fistula tube. Record the conscious blood pressure (BP),blood pressure variability (BPV) and arterial baroreflex sensitivity (ABR) of 1 hour before used durg and 4 hours after.Part two40 rats were randomized into 5 groups: Salvia miltiorrhiza Bge f alba.C.Y.Wu. etH. W. Li high middle and low groups, SHR group and olmesartan(Olm) group. Normal Wistar-Kyoto (WKY) rats were used as negative control. Salvia miltiorrhiza Bge f alba.C.Y.Wu. etH. W. Li and Olm were intragastric administration to rats everyday. The dosage were 15mg/kg.d(Olm),1.5g/kg.d(SM.L),14g/kg.d(SM.H).16 weeks later record conscious blood pressure (BP), blood pressure variability (BPV) and arterial barorecptor reflex (ABR) of 24 hours. left ventricle histological section were disposal by HE stain and VG stain. The left ventricle histological section were test Left ventricular mass index (LVMI), Transdiameter of cardiomyocyte (TDM), Cardiomyocyte area (CA),Collagen volume fraction (CVF), Perivascular circumferentiall area(PVCA). ET,PGI2,TXA2,vWF,TGF-?1,NO,SOD,cNOS of blood plasma were measured. Deploy the technology of TUNEL to test cardiac myocyte apoptosis index(CMAI), smooth muscle cell apoptosis index(SMCAI), Renal Apoptosis Index(RAI).Results1 Compared with WKY rats, SHR exhibited one hand higher BP, LVMI, LVT/RVT, TDM, CA, CVF,PVCA,TXA2,ET,vWF,TGF-?1levels the other hand lower rRPI, RCT/RMT, PGI2, NO, SOD, cNOS,SMCAI levels(P <0.01).2 After Administer single-dose of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li for 4 hours,the systotic blood pressure and BPV in the SHR group was higher than that of WKY group(P <0.01). The BRS was lower than in the WKY group. The systotic blood pressure in the Salvia miltiorrhiza Bge f alba.C.Y.Wu. etH.W.Li groups were almost the same as that in the SHR. The BPV of Salvia miltiorrhiza Bge f alba.C.Y.Wu. etH.W.Li was lower than that in the SHR rats(P <0.05), still higher than that in the WKY rats. The BRS was higher than in the SHR, still lower than in the WKY rats(P <0.05).3 After prolonged Administer of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li for 16 weeks,the systotic blood pressure and BPV in the SHR group was higher than that of WKY group(P <0.01). The BRS was higher than in the SHR, still lower than in the WKY rats (P <0.05).4 After prolonged Administer of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li for 16 weeks, the score of TOD of the SM.H and Olm groups were much lower than the SHR group(P <0.05). The levels of PGI2,NO,SOD,cNOS on SM.H and Olm groups were much higher than the SHR group (P <0.05). The levels of TXA2,ET,vWF,TGF-?1on SM.H and Olm groups were much lower than the SHR group (P <0.05). to compared with SHR group, CMAI,RAI of SM.H and Olm groups were much lower, SM.H and Olm groups of SMCAI were much higher(P <0. 05和0. 01).Conclusion:1 the spontaneous hypertensive rat target organ damage were related to the increasing of BP and BPV2 There was no significant depression of blood pressure after single-dose and prolonged Administer of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li, but Obviously degrade BPV of SHR. The mechanism may be related to improve arterial baroreflex sensitivity3 prolonged Administer of aqueous extract from Salvia miltiorrhiza Bge.f.alba.C.Y.Wu.et H·W·Li, can play a protection of SHR target organ damage. The mechanism may be related to decrease the BPV, relieve the damage of endothelial cell, regulating apoptosis in target organ.
