Objective: To observe the effects of Heat-reinforcing Needling Method on expressions of Atp5 O, Atpase, Sdh m RNA in muscles around ankles of rheumatoid arthritis rats of wind-cold-damp retention, and to explore the molecular mechanisms of Heat-reinforcing Needling Method for “ heat producing ”.Method:Ten rats were selected from forty female SD rats as normal control(N=10), and the rest rats were used to establish rheumatoid arthritis models by Freund’s complete adjuvant and Wind-cold-damp stimulates. Rats, affirmed the model, were divided into three groups randomly. They were model control(N=9), uniform reinforcing-reducing group(N=10),heat-reinforcing needling group(N=10). Rats in the normal group and the model group were just given clutch and bundle without acupuncture. Rats in the uniform reinforcing-reducing group were given uniform reinforcing-reducing needing at Zusanli(ST36), Sanyingjiao(SP6),Guanyuan(RN4) and joint local points, keeping the stimulation 1 minute. Rats in the heat-reinforcing needling group were given heat-reinforcing needling at Zusanli(ST36),Sanyingjiao(SP6), Guanyuan(RN4), but joint local points were given uniform reinforcing-reducing method. Retaining 30 minutes each day, once a day for 7 days. After the treatment, assessed arthritis index and measured the right toe thickness, then sacrificed rats and peeled off ankle muscles and synovial tissue, the real-time quantitative PCR methods were applied to detect expressions of Atp5 O, Atpase, Sdh m RNA in ankle muscle tissue,synovial tissue were amputated for histopathological analysis.Result:1 the effect of heat-reinforcing needling on the rats’ inflammatory integral scoreThe rats’ inflammatory integral score of model group was significantly severer than those of the uniform reinforcing-reducing group and heat-reinforcing group(P<0.05). After treatment, the scores in uniform reinforcing-reducing group and heat-reinforcing group were obviously lower than that of the model group(all P<0.05), and the change of the heat-reinforcing group was much clear than that of uniform reinforcing-reducing group(P<0.05).2 the effect of heat-reinforcing needling on the rats’ toe thicknessThe rats’ right toe thickness in model group was obviously increased compared with that of the normal group(P<0.05). After treatment, rats’ right toe thickness both in the heat-reinforcing needling group and uniform reinforcing-reducing group were significantly lower than that of the model group(P<0.05), and the change of the former group is better than that of the later one(P<0.05).3 The effect of heat-reinforcing needling on the pathomorphology of the synovial membrane of RA rats.After treatment, synovial tissue structure of rats of the normal group was normal; that in the model group was anomalous, we observed many synovial B lymphocytes, fibrous connective tissue proliferated, and new neovascularization; In addition, the synovial membrane of rats in heat-reinforcing needling group and uniform reinforcing-reducing group were improved than that of model group, and the former group is better than the later one.4 The effect of heat-reinforcing needling on the expressions of Atp5 O, Atpase, Sdh m RNA in rats’ kneel joint muscle.Compared with the normal group, the expressions of Atp5 O m RNA, Sdh m RNA in model group rats were much lower than those of normal group( all P<0.05), but the expression of Atpase m RNA had no significant difference among two groups(P>0.05).Compared with the model group, the expression of Atpase m RNA in uniform reinforcing-reducing group was obviously enhanced(P<0.05), while the expressions of Atp5 O m RNA and Sdh m RNA in uniform reinforcing-reducing group had no obvious changes( all P>0.05); the expressions of Atp5 O m RNA, Atpase m RNA and Sdh m RNA in heat-reinforcing needling group were obviously enhanced(all P<0.05).Compared with the uniform reinforcing-reducing group, the expression of Atp5 O m RNA,Atpase m RNA and Sdh m RNA were all obviously increased in the heat-reinforcing needling group(all P<0.05).Conclusion:(1) The heat-reinforcing needling method could obviously raise the expressions of Atp5 O m RNA, Atpase m RNA and Sdh m RNA in RA rats’ ankle muscle tissue.(2) The heat-reinforcing needling method could obviously improve the inflammatory integral score and the toe thickness of RA rats.(3) Up-regulation the energy metabolism related upstream genes Atp5 O, Atpase and Sdh m RNA expression was one of the channel of “ heat production ” for Heat-reinforcing Needling method.
