Objection: To investigate the expression and interaction relationship of Caveolin-1 and GPR30 in human papillary thyroid carcinoma(PTC) patients, and assess the association with tumor invasion and migration.Methods: Immunohistochemistry was used to analyze the expression of Caveolin-1 and GPR30 in 76 PTC specimens, and the correlations with clinicopathological indicators. We constructed Caveolin-1-si RNA, Caveolin-1-pcDNA3.1(+) plasmids, transfected the plasmids to K1 and BCPAP cell lines of PTC, detected mRNA and protein levels of GPR30 by RT-PCR and Western blot respectively, and tested the invasion and metastasis ability of each cell line’s. Using GPR30 selective agonist G1 and GPR30 selective antagonist G15 treated K1 and BCPAP cells, and detected mRNA and protein levels of Caveolin-1 by RT-PCR and Western blot, respectively.Results: Immunohistochemical results showing that in 76 cases of PTC, the positive expression rates of Caveolin-1 and GPR30 were 9.21% and 80.26%, respectively. The positive expression rate of Caveolin-1 in PTC was markedly lower than in normal tissues, and the positive expression rate of GPR30 in PTC was higher than in normal tissues, the difference had statistically significant( P<0.001). Caveolin-1, GPR30 expression had significant correlations with TNM stages(P=0.005,P<0.001, respectively) and with cervical lymph node metastasis( P≤0.001 for both). Meanwhile, the expression of Caveolin-1 had a negative correlation with GPR30( rs=-0.528, P<0.001). Silence Caveolin-1 gene expression up-related the mRNA and protein expression levels of GPR30, promoted the ability of cells wearing of membrane and migration. Over expression of GPR30 inhibited the mRNA and protein expression levels of Caveolin-1.Conclusion: All above demonstrated that there may be interaction relationship between Caveolin-1 and GPR30 that affect the invasion and metastasis of tumor cells in PTC. Caveolin-1 and GPR30 may be used as a significant prognostic in PTC.
