Pericarpium Citri Reticulatae from Guangdong(Guang Chenpi in chinese) is the top of ten famous-region drugs in Guangdong province. The quality goods are the drying ripe pericarp of Citrus reticulata’chachi’,it is mainly planted in Xinhui, Guangdong province.Guang Chenpi has a high medicinal value,like regulating vital energy and invigorating the spleen, harmonizing the stomach and stop vomiting, drying the wetness-evil and eliminating phlegm. Pharmaco-research showed that polymethoxylated flavones exhibited diverse pharmaco activities,as anticancer, antioxidant, anti-inflammatory, antimutagenic, cardio etc.To identify the genuine phytochemical constituents and control the quality of the famous-region drug, phytochemical studies focused on the lipophilic components were undertaken firstly which lay the first stone for pharmacological and drug efficacy test and quality standard research and the standard of quality control for Guang Chenpi was set up.Traditional methods for the extraction and isolation were applied. As the result, seven compounds were obtained from the petroleum ether part extracted from ethanol extract of this medical material,and the structures of seven compouds were elucidated by spectral(1H-NMR,13C-NMR, MS) and chemical methods.They are o-montanic acid、β-sitosterol、5-hydroxy-6,7,8,3’,4’-pentamethoxyflavone、tangeretin、nobiletin、daucosterol、hesperidin respectively.Furthermore, three bioactive flavones (hesperidin, nobiletin, tangeretin) were selected as chemical markers, according to the results of the separation and isolation experiments.Applying a high performance silica gel G plate, first using acetic ether-methanol-water(100:17:13),then using the upper layer of methylbenzene-acetic ether-formic acid-water (20:10:1:1)as developing solvent. The plate was scanned at wavelengths ofλs=265nm,λR=600nm. A simple, precise, accurate and reliable analytical method for the simultaneous determination of the three flavones was developed.Thus it is an important reference for total quality control of Guang Chenpi.
