Study On Ultramicro-Buyang Huanwu Decoction On Cerebral Infarction Convalescent Clinical Curative Effect And Effect Of Neural Stem Cells Apoptosis In Rat Focal Cerebral Ischemia

Objectives:Study on ultramicro-Buyang Huanwu decoction on cerebral infarction convalescent clinical curative effect and effect of neural stem cells apoptosis in rat Focal cerebral ischemia, To provide scientific bases for clinical treatment.Methods:1.60 case of patients with cerebral infarction recovery from hunan 2008.6 to 2008.11 were selected and divided into two groups randomly (treatment group and control group),and treated with Buyang Huanwu and ultramicro-Buyang Huanwu respectively for 4 weeks.lt was designed to observe the changes of neurologic impairment scores,stroke scores,Advese reaction was also obversed.2. Hippocampal stem cells was mechanical separated from newborn rats SD,quantum dot labeled was used to observe the conditions of their labels.3. Rat focal cerebral ischemia was induced by MCAO, quantum-labeled neural stem cells were transplantated into brain through different channels,survve and differentiation of the transplanted cells and animal deaths were observed.Rat model of focal cerebral ischemia was induced, rats were randomly divided into 5 groups including model group,blank transplantation group,simply transplantation group,Chinese herbs plused stem cell transplantation group and ultramicro Chinese herbs plused stem cell transplantation group,rat were given different treatment. Evaluation of nerve function,differentiation and cell survival was observed in 7 days,14days and 28 days after transplantation.tested levels of Bag-1、Bax、Bcl-2 gene expression.Results:1. Clinical efficacy of the two groups rank sum test for analysis, results show that the difference between the two groups was not significant (P> 0.05); of two cases of curative effect of TCM Syndrome Analysis of rank sum test, the difference was statistically significant (P<0.05), indicating that symptom efficacy of TCM treatment group than the control group.2. The improvement of neal udystrophyrologic impairment scores in treatment group is statistical significance,But the difference between two groups was no statistical significance. (P> 0.05) The improvement of stroke scores in treatment group,was better than in control group,The difference between two groups was statistical significance (P< 0.05)3. In the course of treatment,no adverse reaction was been found in both.4. Neural stem cells can be labeled by quantum dot,and can be integrated into cell progeny.5. All groups exist neurologic impairment after cerebral ischemia.The improvement of neurologic impairment scores.there was no statistical different Neural stem cell transplantation group than in model group and blank group were transplanted, at 14,28 days, there was a significant difference (P<0.05); United Buyanghuanwu Decoction of neural stem cell transplantation, AMD United Buyanghuanwu soup than the neural stem cell transplantation pure neural stem cell transplantation group, the three groups at 14,28 days there was a significant difference (P<0.05); the model group and the blank transplantation group differences at each time point was not obvious.6. Bag-1, Bcl-2 gene in normal brain tissue has a low level of expression, the expression of cerebral ischemia at the level of increase in stem cell transplantation group Bag-1, Bcl-2 gene expression in model group than the increase in cell transplantation group combined traditional Chinese medicine, Advanced Micro Devices combined cell transplantation group of Chinese medicine can increase Bag-1, Bcl-2 gene expression,7,14,28 d in the three groups there was a significant difference, P<0.05 or P<0.01.7. Bax gene in normal brain tissue has a low level of expression, the expression level after cerebral ischemia enhanced stem cell transplantation group the level of Bax gene expression in model group than the increase in cell transplantation group combined traditional Chinese medicine, Chinese medicine combined cell transplantation AMD group Bax gene expression can be reduced, 7,14,28 d in the three groups there was a significant difference, P<0.05 or P <0.01.Conclusions:1. Buyanghuanwu Decoction can improve the patient’s symptoms, the treatment of cerebral infarction are safe and effective recovery of the ideal of non-toxic drug reaction.2. The vast majority of neural stem cells can be marked quantum dots.3. There is a certain amount of normal brain of the Bag-1, Bcl-2 gene expression, Buyanghuanwu soup can promote post-ischemic brain injury Bag-1, Bcl-2 expression.4. There is a certain amount of normal brain gene expression of Bax, soup Buyanghuanwu resistant to injury after cerebral ischemia the expression of Bax.5. Micronization can improve the utilization of traditional Chinese medicine herbs,1/3 the volume of powder and pieces of the Bag-1, Bax, Bcl-2 gene that is affected with a considerable amount of the role of traditional pieces. Pieces of the whole volume of powder is superior to the traditional pieces total.
