University teachers are engineers of higher education and the human soul. Good teacher-student relationship is the precondition to create the best students. Good teacher-student relationship can not only improve the level of education of the whole teaching, but also promote the harmonious development of the student’s body and mind. In this study, college students are subjects, and establish the "university student-teacher’s alienation Questionnaire" through literature and open questionnaire, and carry out a questionnaire survey to know the situation of college student-teacher’s alienation, self-differentiation, self-esteem, and the relationships among them were discussed also. We can understand the antecedents and mediating variables of student-teacher’s alienation in depth. In this way we can reduce the alienation between teachers and students, and provide a perspective of building a harmonious campus.The main conclusions are as follows:(1) The "university student-teacher’s alienation Questionnaire" has a good reliability and validity, a total of 21 questions. It is divided four factors as sense of escaping, sense of meaningless, sense of strangeness and sense of powerlessness.(2) University teacher-student’s alienation is low, indicating a good teacher-student relationship. And high alienated individuals are only 19% of the total. And the alienation of undergraduates (except freshman) are generally higher than graduates’. The students who are cadres have less alienation than those who not. There is a big difference in the profession and gender to some factors.(3) University students have a low level of self-differentiation, but have a very high self-esteem.(4) Explicit self-esteem has a negative effect on predicting the student-teacher’s alienation, while implicit self-esteem can not directly predict the student-teacher’s alienation, and self-differentiation has a positive effect on predicting the student-teacher’s alienation.(5) The relationship between explicit self-esteem and the student-teacher’s alienation is partially mediated by self-differentiation, and the relationship between implicit self-esteem and the student-teacher’s alienation is fully mediated by self-differentiation. And the influence self-differentiation to the student-teacher’s alienation is adjusted by esteem structure in a certain role.
