Purpose:It has been reported that adipose tissue is an endocrine organ, the adipose tissue can secrete many types of adipokines to improve lipid metabolism. Therefore it can reduce the incidence of obesity, type II Diabetes (T2D) and cardiovascular disease, etc. We try to apply 60%-70%V02max intensity exercise training in SD rats, to find the effects on the expression of FNDC5/Irisin protein in white adipose tissue and muscles of obese rats. Object and Method:(1) There were 80 Three-week-old male SD rats.60 SD rats were selected as obesity modeling group randomly, also as high fat group (HFG).20 SD rats were selected as control group (CG). HFG rats were fed on high fat diet (45% fat content percentage), CG rats were fed on normal fat diet(10% fat content percentage). The feeding time were 8 weeks. Examine 80 rats’original weight and body length. Every weekend examine rats’weight and body length in 8-week feeding time to calculate Lee’s index. (2)There were 20 HFG rats becoming obese rats, then divide them into two groups:obese control group (OCG) and obese exercise group (OEG). Every group was 10 rats. Divide 20 control group rats into two groups:control secondary group (CG) and control exercise group (EG). Every group is 10 rats. Exercise group rats performed 8-week treadmill training, the training intensity is 60%-70%V02max,5 days a week and 1 hour per day. The control group rats didn’t do any training. During 8-week exercise training period, every weekend records rats’weight and body length, at 15th week examine 20 rats’body composition (4 groups,5 per group). At the end of 16th week, all rats were anesthetized and sacrificed. Then we get rats’blood in rats’ abdominal aorta, white adipose tissue (WAT), brown adipose tissue (BAT), Gastrocnemius and Soleus. Blood were centrifuged and extract the supernatant. Aliquoting and put the samples into -80℃ refrigerator to prepare the test the blood indexes. Weigh the contents of WAT and BAT. Aliquoting and put the samples into -80℃ refrigerator to prepare the western blot (WB). Results:(1) It was successful to build the obese rats’ model in 8 weeks. HFG rats’weight were significantly higher than CG rats and exceeded 18.4%, p<0.01. HFG rats’Lee’s indexes were significantly higher than CG rats, p<0.05. (2) 8-week 60%-70%V02max treadmill training significantly improved FNDC5/Irisin protein expression of epididymal WAT, kidney WAT in OEG rats. The UCP-1 protein expression of BAT was siginificantly increased in OEG rats. It indicanted that exercise could stimulate WAT’s transition to BAT and in result to change the obese rats’body fat percentage. Conclusion:(1) Three-week-old weaning rats were fed on high fat diet for 8 weeks and it succeeded to build the obese rats’model. (2) 8-week 60%-70%V02max intensity exercise interfere could increase FNDC5/Irisin protein expression of epididymal WAT and kidney WAT in obese rats, which resulted in WAT’s transition to BAT and improved the obese rats’ body fat levels.
