ObjectiveTo investigate the influence of exogenous ZAG and aerobic exercise on theexpression of ZAG mRNA and UCP-1mRNA in nutritive obesity mice, trying tofinding the effect of exogenous ZAG on weight loss, and further exploring themechanism of exercise on weight loss.Materials and Methods140KM male mice weighing30g were randomly divided into two groups: normalfood group(C,n=20) and high-fat group(n=120). It is suggested that a mice in high-fatgroup whose weight higher than normal control group mice for20%can be identifiedas obesity. Obesity mice were randomly divided into5groups: obesity group (D, n=9),placebo group (P, n=9), ZAG group (Z, n=9), ZAG+exercise group (Z+E, n=9) andexercise group(E, n=9). Injection design: P group receiving10μl lipidosome and300μl OPTI-MEM mixture in tail vein for twice each week,8weeks. Z group and Z+Egroup were treated with injection of5μg ZAG plasmid,10μl lipidosome and300μlOPTI-MEM mixture via caudal vein for twice each week,8weeks. Exercise program:Z+E group and E group were given a daily load of10m/min treadmill exercise at60min/d,5d/wk for4weeks and12m/min for another4weeks. Body weight wasrecorded every three day. After8week experiment, the gain of weight, the weight ofvisceral fat and brown adipose tissue (BAT) in scapula were procured. The serum wasseparated for detecting TG, TC, LDL and HDL. The expression level of ZAG andUCP-1mRNA in BAT was measured by fluorescence quantitative PCR. The characterof brown adipose tissue was observed by HE stained.Results1) After8weeks interventions, the weight gain and the relative weight of visceral fatof Z group were significantly higher than those of D group. The increased ofweight of E group was significantly higher than that in mice at C group (p<0.01),while there was no significantly different between E group and D group. Therelative weight of visceral fat of E group was significantly lower than that in miceat D group (p<0.05), while there was no significantly different between E groupand C group. The increased of weight of Z+E group was significantly lower thanthat in mice at D group (p<0.01) and E group (p<0.05), while there was nosignificant difference in other groups. The relative weight of visceral fat of Z+Egroup was significantly lower than that in mice at D group (p<0.01), while therewas no significant difference in other groups.2) The concentrations of HDL in Z group was significantly higher than C group(p<0.05) and D group (p<0.05), while its TC was significantly lower than that inmice at these two groups (p<0.01,p<0.01). The concentrations of TG and LDLwere no significant difference between Z group, C group and D group. The serumcontent of TG in mice at E group was significantly lower than that in mice at Cgroup (p<0.05) and D group (p<0.05), while the serum content of TC, HDL and LDL were no significant different between in mice at E group, C group and Dgroup. The serum content of TG and LDL were no significant different between inmice at Z+E group and other groups.3) The expression of ZAG mRNA in Z group was significantly higher than that inmice at D group (p<0.01), and there was no significant difference between Zgroup and C group. The expression of UCP-1mRNA in Z group was significantlyhigher than that in mice at C group and D group. The expression of ZAG mRNAin mice at E group was significantly higher than that in mice at D group (p<0.05),but significantly lower than that in mice at C group (p<0.01). the expression ofUCP-1mRNA in mice at E group was significantly higher than that in mice at Dgroup (p<0.05), and there was no different between E group and C group.4) The expression level of ZAG mRNA in the adipose tissue showed significantpositive correlation with the level of UCP-1mRNA (P<0.01).ConclusionsExogenous ZAG and aerobic exercise can increased the level of UCP-1mRNA,reduce the amount of visceral fat, improve obesity by up regulating the expression ofZAG mRNA in the brown adipose tissue.
