Objective: Autophagy is a highly conserved mechanism of self-phagocytosis,which is closely related to the occurrence of obesity.However,the mechanism of obesity and exercise training induced specific changes in autophagy may be helpful for the treatment of obesity-related diseases.Therefore,the aim of this study is to observe the effects of moderate intensity exercise on the expression of autophagy-related proteins in different adipose tissues of obese rats,so as to provide a theoretical basis for understanding obesity and lose weight.Methods:Twenty obese rats were randomly divided into four groups: the control sedentary group(C),thecontrol exercise group(CE),obese sedentary group(O)and obese exercise group(OE).Exercise group rats were subjected to moderate intensity exercise for 8 weeks,5 days per week,1 h per day.48 hours after the last exercise drawn,respectively,to take testicular white fat,subcutaneous white fat and scapular brown adipose tissue,stored at-80?C refrigerator.The protein expression of LC3 II / I,ATG7,ATG5,ATG12 and ATG12-ATG5 were measured by Western Blot.Result:1.The protein expression of LC3 II / I,ATG7,ATG5,ATG12 and ATG12-ATG5 in obese sedentary group were significantly increased(p <0.05)in subcutaneous white fat compared with the control sedentary group,(P <0.05),while the expression of LC3 II / I and ATG7 in the obese exercise group was significantly lower than that the obese sedentary group(p <0.05).ATG5 and ATG12-ATG5 in the tests were significantly increased(p <0.05)compared with the standard quiescent group,while ATG7 and ATG5 were significantly higher in the obese group ATG5,ATG5 and ATG5-ATG12 in the obese exercise group were significantly lower than those in the obese group(p <0.05),while ATG12-ATG5 was significantly higher(p <0.01).ATG5,ATG12 and ATG12-ATG5 were significantly increased(p <0.05)in the standard quiet group compared with the standard resting group.LC3 II / I and ATG12-ATG5 in the obese quiet group had ATG7,ATG12 and ATG12-ATG5 in the obese exercise group were significantly higher than those in the obesesedentary group(p <0.05),but there was no significant difference(P> 0.05).Conclusion:1.High-fat diet induced obesity can enhance the expression of LC3 ubiquitin-like conjugation system related proteins of white adipose tissue,but it can inhibit the brown adipose tissues' s expression.2.8 weeks of moderate-intensity exercise can inhibit the expression of LC3 ubiquitin-like conjugation system related proteins of obese rats with white fat,but can enhance the expression of brown adipose tissue.
