Coordination chemistry is one of the frontier subjects of modern chemistry,whose researches focus on the design, synthesis and related physical, chemical,biological properties of functional complexes. Initial screened, compared anddiscussed the anti-tumor, anti-bacterial and other biological activityof the novelligands and complexes, including the possible configurations-effect relationship. Inaddition, study the antitumor mechanism of ligands and complexes throughanti-oxidation, cell cycle and the apoptosis inducing effect. This topic research hasimportant theoretical significance to realized the guiding role of bioinorganicchemistry theory in new drugs synthesis. This research has important practical valueto broaden the biological application of schiff base complexes and bioactiveamine.The main contents of this study were stated as following:Eighteen kinds of unreported schiff base ligands and metal complexes weresynthesized by solvothermal method.(1) Six novelschiff base ligands:1-phenyl-3-methyl-4-(4-toluoyl)-pyrazolone-5(A1) and p-aminophenol(L11), p-nitroaniline(L12),?-alanine(L13) schiff bases;1-phenyl-3-methyl-4-(?-furoyl)-pyrazolone-5(A2) andp-aminophenol(L21) schiff base;1-phenyl-3-methyl-4-(4-chlorobenzene formyl)-pyrazolone-5(A3) and ?-alanine(L31), p-aminophenol(L32)schiff bases.(2) Threenovelschiff basecomplexes: N-(1-phenyl-3-methyl-4-(4-toluoyl)-pyrazolone-5)-?-alanine schiff base Cu(CuL13), Ni(NiL13) complexes; N-(1-phenyl-3-methyl-4-(4-chlorobenzene formyl)-pyrazolone-5)-?-alanine schiff base Cu(CuL31)complexes.(3) Nine novel4-acylpyrazolone complexes:1-phenyl-3-methyl-4-(4-toluoyl)-pyrazolone-5Cu(CuA1), Ni(NiA1), Co(CoA1), Zn(ZnA1)complexes;1-phenyl-3-methyl-4-(4-chlorobenzene formyl)-pyrazolone-5Cu(CuA3),N i (N i A3), C o (C o A3), Z n (Z n A3) c o m p l e x e s;1-p h e n y l-3-m e t h y l-4-(3-toluoyl)-pyrazolone-5Cu(CuA4) complexes.All the schiff bases and their transitional metallic complexes have beencharacterized by elemental analyses, molar conductance, IR spectra, UV spectra,TG-DTA, XRD and cyclovoltammetry to determine their composition and structure.Study the anti-proliferative effect of L13, L31, CuL13, NiL13and CuL31on human colon cancer cell(LoVo). The results showed that all of the ligand and complexes haveobvious anti-tumor activity and the dose-response relationship was a significantpositive correlation.The CuL13has the strongest anti-tumor activity in vitro.Theanti-bacterialresearch showed that, all of the ligands and complexes have varyingdegrees anti-bacterial effect againstGram-negative bacteria (Escherichia coliATCC11229) and Gram-positive bacteria (Staphylococcus aureusATCC6358P). Thestudy on the mechanism of anti-tumor showed that,the complexes achieve theanti-tumor activity is not through anti-oxidant, but arrest the cell cycle at G2M andapoptosis-inducing effect.Through the structure-activity relationship analysis, wefound that the stronger the electron-withdrawing of the substituents of4-benzoyl ofpyrazolone ring is, containing more hetero atoms, the better the biological activity ofthe metal ions is,the smaller the steric hindrance is, the complexe has strongeranti-tumor activity.
