Osmunda japonica Thunb is a herbaceos perennial fern with considerable nutritional?medicinal and economic values,which belongs to Pteridophyta,Protoleptosporangiopsida,Osmundales,Osmundaceae,Osmunda.Although the semi-processed products of osmund could create certain economic benefits,there were still some disadvantages such as product structure single?strong seasonality?aging?basic theory imperfect?further study lag.As a result,both of the product added value and resource utilization were quite low.This study was to do some extension and supplement on the basic research of osmund,attempting to further fill the blanks of the basic theory and provide certain theoretical reference for its depth utilization and development.The specific research contents were shown as follows:1.Optimization of Extraction Technology for Soluble Dietary Fiber from Dried OsmundResponse surface methodology was applied to optimize the technological parameters of the ultrasonic-assisted alkaline extraction of soluble dietary fiber from dried osmund.Effects of four independent variables including solid-liquid ratio,extraction temperature,ultrasonic time and ultrasonic power on the yield of soluble dietary fiber were significant by single-factor experiment.A quadratic regression mathematical model was established by Central-Composite experimental design and analyzed by response surface methodology,meanwhile the influences of independent variables and their interactions on the response value were discussed.The optimal technological parameters were determined as follows: alkaline concentration 0.02g/m L,extraction time 90 min,solid-liquid ratio1.0g/23 m L,extraction temperature54?,ultrasonic time 25 min and ultrasonic power380 W.Under the optimal extraction conditions,the yield of soluble dietary fiber was up to38.39%.Water holding capacity was 8.82g/g,swelling capacity was 9.35 m L/g,which showed that soluble dietary fiber from dried osmund had fine physicochemical properties.2.Analysis of Amino Acids Composition and Nutritional Value Evaluation of Dried OsmundTo systematically analyze and evaluate amino acids of dried osmund,17 kinds of amino acids were derivatized with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate and then were analyzed by ultra performance liquid chromatography.L-tryptophan was quantitatively analyzed by ultraviolet spectrophotometric.Nutritional value evaluation of dried osmund protein was synthetically studied by methods of ratio coefficient of amino acid and fuzzy discernment.The results showed that the sample had 18 kinds of amino acids.The content of the total amino acids was up to 697.93mg/gprotein.The proportion of essential amino acids in total amino acids was 0.45,and the proportion of essential amino acids in non-essential amino acids was 0.83,which was higher than that of FAO/WHO pattern recommended.The content of crude protein was 12.09%.The contents of flavor amino acids?sweet amino acids?aromatic amino acids and medical amino acids were23.99%?22.16%?12.46% and 63.97% respectively.All kinds of human essential amino acids kept such a balanced proportion that they were close to the FAO/WHO standard pattern.Among all human essential amino acids,the first limited amino acid was isoleucine.3.Evaluation of Measurement Uncertainty for Determination of Dried Osmund Total Ash and Acid-insoluble AshTo evaluate the measurement uncertainty of total ash and acid-insoluble ash of dried osmund,the indexes were tested according to the gravimetric method,meanwhile,a mathematical model was established on the basis of assay method,then the main sources of uncertainty were analyzed,and the measurement uncertainty was calculated and assessed in the light of JJF1059.1-2012<Uncertainty Assessment and Expression>.The content and expanded uncertainty of dried osmund ash and acid insoluble-ash were?6.5440±0.080?%and?0.5000±0.035?% respectively.4.Study on the Extraction Condition and Antioxidition of Polyphenol from Dried OsmundThis study was to explore optimal extraction conditions and antioxidant activity of polyphenol from dried osmund.The polyphenols was extracted by ultrasound-assisted ethanol extraction method and quantified by Folin-Ciocalteu method.The orthogonal test was adopted and the factors influencing the process parameters was determined.The antioxidant activity of polyphenol from dried osmund was evaluated through mesuring reduction force and detecting the scavenging rate of 1,1-diphenyl-2-phenyl free radical?DPPH·?,hydroxyl free radical?·OH?and ultra oxygen anion free radical?O2-·?,compared with vitamin C.The results showed as follows,the optimal technological parameters were ethanol concentration 80%,solid-liquid ratio 1:20?g/m L?,ultrasonic time40 min and ultrasonic power 100%.Under the optimal extraction conditions,the reduction force which was expressed as ?A was 0.50,the half elimination concentration of1,1-diphenyl-2-phenyl free radical?DPPH·?,hydroxyl free radical?·OH?and ultra oxygen anion free radical?O2-·?was 6.1372?g/m L?1.8522?g/m L?0.425 mg/m L respectively,which was similar to that of vitamin C.
