Garlic is a liliaceae allium plants and has been used as food and medicine for thousands of years.With the research and application of garlic medical care value,garlic gets more and more the favour of people.Alliin is a kind of flavor precursors in garlic and has many roles,such as antibacterial,antiviral,scavenging free radicals,improving immunity,anti-aging,antineoplastic,reducing blood fat,degrading cholesterol and prevention of atherosclerosis.When garlic is suffered from exogenic action,exogenic action will rupture and alliin will react with allinase of vacuoles to generate allicin and other substances.Allicin also has antibacterial and antitumor activity but it is unstable and easy to degrade,therefore,the activity of the original will also be lost.So,this study was conducted to discuss to use CTAB reverse micelle system to extract alliin from garlic and optimize the process conditions.The main contents are as follows:(1)The suitable extraction method of alliin in garlic powder was obtained according to the exploration for the facters of HPLC,the result was: chromatographic column: Ufavour ODS-C18(4.6×250 mm,5?m),wavelength: 210 nm,mobile phase: 10% methyl alcohol and 90% water,flow rate: 0.3 m L/min(0-15min),injection volume: 20?L,column temperature: 35?.Under the chromatographic condition described above,the peak occured at about 10.918 min was considered as the signal of alliin,and the degree of separation all above 1.0,which illustrated a better separating effect.In the meantime,drawing a alliin standard curve according to the gradient measurement for alliin accustandard making use of the chromatographic condition described above.(2)Optimizing the extraction condition of alliin using the mixed system of CTAB/n-caprylic alcohol/n-heptane,various facters included extraction time ?concentration of CTAB?temperature?concentration of KCl?pH?adding amount of garlic powder....have graet influence on the alliin extraction effect,according to the single factor experiment and orthogonal experiment respectively,obtainig the extraction condition which results the highest extractability.On the basis of the date obtained from orthogonal experiment,the best extraction condition of the mixed system was: concentration of CTAB was 0.07mol/L,PH of buffer solution was 7.0,concentration of KCl was 0.3mol/L,extraction time 30 min.Optimizing reverse extraction rate was 1.62%.(3)By changing the conditions of the aqueous phase of back-extraction to investigate the influence of different conditions on the efficiency of back-extraction.Such as the ion concentration in the aqueous phase,the pH of the aqueous phases and the ratio between the organic phase and water phase.Based on the orthogonal experiment data,the best reverse Extraction conditions were the pH of the aqueous phases was 5.0,the concentration of KI was 0.8 mol/L,the ratio between the organic phase and water phase was 1:3.The best efficience of back-extraction was 1.26%?Based on the orthogonal experiment getting that the influence of different factors on the alliin extraction yield effect is: the ion concentration> the ratio between the organic phase and water phase> the pH of the aqueous phases.
