This research adopts the cordyceps sinensis which is the real asexual type species as our fermentation strains. During the research, cordyceps mycelium fermentation technology was explored and optimized.We completed the laboratory stage amplification of mycelium fermentation step by step,and analyzed effective constituents, and completed the pilot stage to carry out the subject, the main works are as follows:We completed laboratory stage 5L,30 L,300L fermentation scale amplification process step by step, and the indicators have reached the industry standard.By improving the culture medium the way of sample processing, we doubled the content of adenosine in the mycelium, the highest level was 724 mg/100 g, and the average level was 400-500 mg /100 g,, the highest dry weight is 23 g/L,and the average dry weight was 11 to 15g/L, the content of mannitol was 9-10g/100g.We detected some new effective components analysis in cordyceps mycelia by using UPLC-MS such as GABA/SAM/ergosterol/cordycepin /guanosine.We carried out the quantitative analysis results.In our homemade fungus powder,the content of GABA is 800-1000 mg/100 g, the optimized SAM content can be up to 686.27 mg/100 g, the ergosterol content was up to 360 mg/100 g, the highest cordycepin content was 23 mg/100 g, guanosine content was 790.94 mg/100 g, uridine content was 930.37 mg/100 g, hypoxanthine content was 123.54 mg/100g;We studied the metabolic fingerprint and compared our fermentation mycelia with natural cordyceps and commercial fungus powder in the effective ingredients, quantitative analysis of the results proved that our fungus powder has advantage in similar products;We also determined the content of heavy metal and microorganism toxin in mycelium powder, the results showed that our powder does not contain heavy metals and microbial toxins;We went to Henan Province to work for the pilot magnification in this cooperation project. We completed 50L of tank and 5T of tank for cordyceps mycelia fermentation.We finally got 19.5 kg of powder, fungus powder adenosine contont in the powder was above 300 mg/100 g, mannitol content is more than 7 g/100 g.We shorten the fermentation period for 4 days,thus improving the production efficiency.
