Lactic acid bacteria have excellent efficacy,functional foods with plenty of lactic acid bacteria help to maintain intestinal balance,improve immunity,prevent cancer,lower cholesterol and improve constipation and diarrhea other functions.However,lactic acid bacteria and their effect on human health of the intestines is closely related to the number of viable cells.Gastrointestinal environment in a variety of adverse factors,particularly gastric acid,the number of viable bacteria cause a significant decrease in lactic acid bacteria,which greatly limits the play to its probiotic capabilities..Sodium alginate is the most widely used microcapsule wall material,however,there are some shortcomings,such as high bio-molecule leakage,low mechanical strength,pore bulky and other factors play a negative effect on their prebiotic effect,sodium polyacrylate with a package buried probiotic potential,it is possible to protect the safety of probiotics through the gastrointestinal tract and to assist their colonization in the intestine,it is commonly used in coating materials,food security is relatively high,and more for embedding drugs currently no its probiotic applications in research and reports.For this experiment,the gastrointestinal tract,frozen storage and heat treatment of the environment is not conducive to the survival of lactic acid bacteria,to develop a more effective barrier wall material adverse environment embedding the polymer modification technology applied to the structural design of the wall material,the successful preparation alginate grafted sodium polyacrylate.In this experiment,sodium polyacrylate grafted alginate and sodium alginate composite for the inner wall of the timber,the outer coating made of chitosan microcapsules with alginate microcapsules as a control.Successful grafting determined by IR and NMR detection;viscosity detected by preparing a solution viscosity less than the wall material existing natural polymer material,reducing the difficulty of preparing microcapsules;material added to the prepared culture containing microbial cells It was observed in the growth of bacteria,the bacteria through the normal controls,demonstrating the graft material has no effect on cell growth.This experiment is about the preparation of microcapsules,microcapsules in simulated gastric juice and intestinal juice environment the change of physical properties,the microcapsule embedding lactobacillus MA2 respectively,soaking in simulated gastric juice(pH = 2.0)2 h,Alg/Alg-PAAs microcapsules and Alg microcapsule after 2 h of bacteria live number respectively 9.3 log CFU/mL,7.86 the log CFU/mL,and free bacteria after 2 h of bacteria live number 4.13 log CFU/mL,Alg/Alg-PAAs microcapsule has greatly increased the gastric environment the number of living bacterium.Respectively the embedding the lactobacillus MA2 Alg/Alg-PAAs microcapsules with Alg microcapsule soaked in the simulated intestinal juice after 4 h,the release rate of the latter two times,can see Alg/Alg-PAAs microcapsule can better release in the intestinal tract,smooth the lactic acid bacteria to colonize the intestinal.To erythromycin resistance of GFP gene into MA2 in animal experiments,research the body wall digestion environment protection and promote the activity of lactic acid bacteria probiotics on the surface of intestinal adhesion ability,after oral microcapsule 24 h and 72 h,ileum and colon of rats,extraction of their contents are coated plate count,Alg/Alg-PAAs microcapsules in 24 h after oral lactobacillus MA2 still maintaining high bacteria live.Observed under the condition of refrigeration and heating at the same time under the condition of the susceptibility of microcapsule,can be concluded that the micro capsule can protect the lactic acid bacteria from adverse environmental impact,and Alg/Alg-PAAs microcapsule protective effect is better.In conclusion,the experiment use polymer polymerization method that sodium polyacrylate grafted onto sodium alginate,comparing with sodium alginate greatly improved microcapsules adverse environmental tolerance,so it will lay the foundation for live lactic acid bacteria health care.
